Objective:The study is aim to explore the mechanism of the drug Galangin on RCC 786-Oand caki-1cells. And to provide the relative theory basis for the clinical application.Methods:In the present study, The effect on cell proliferation was examined by CCK-8 assay. Cell cycle was analyzed by flow cytometry. Cell invasion assays and wound healing assay were used to observe galangin on invasion and migration. The expression of ROS、SOD、T-AOC and MDA were detected by western blot test. And the proteins relation to EMT were also detected by the western blot test.Result:1.The results demonstrated that proliferation of 786-0 and caki-1 cells was suppressed when exposed to various doses of galangin.2.The apoptosis induced by galangin was analyzed by flow cytometry. The results showed that galangin could induce apoptosis in a dose-dependent manner. Accumulation of Reactive oxygen species (ROS) is an important contributing factor for 786-0 and caki-1 cells apoptosis. With an increased the levels of SOD and MDA while decreased those of T-AOC.3.Cell scratch test and Transwell test showed that Galangin suppress invasion and migration in renal cell carcinoma cells by suppressing epithelial mesenchymal transition (EMT), with an increased the levels of E-cadherin and decreased those of N-cadherin and vimentin. 100μM Galangin on the cells can produce the maximum inhibitory effects.Conclusion:1.Galangin exerts an anti-proliferative property and dose-dependent character.2.Galangin can induce apoptosis in renal cell carcinoma cells and exert the dose-dependent feature.3. Galangin inhibits renal cell carcinoma invasion and migration4. Galangin may prevent renal cell carcinoma growth, inhibit cell invasion by EMT and induction of cell apoptosis by producing reactive oxygen species.
