| [Objects] Assessing the therapy and effect of tonifying kidney and removing stasis and reducing phlegm on cognitive function and oxidative stress by clinical observation and animal trials. To explore oxidative stress effects cognitive function, and discussing the pathological mechanism of TCM preventing brain aging.[Method]In the part of clinical observation, the level of cognitive function and oxidative stress of84patients of mild cognitive impairment are affected by the therapy of tonifying kidney and removing stasis and reducing phlegm or lifestyle intervention. We use the scale of MMSE and MoCA to compare the change of patients before and after the intervention. And detect the level of SOD, GSH-Px and MDA in fast venous blood in the morning.In the part of animal trial, ApoE knockout mice have been divided into four groups as the treatment group and model group of senior and the treatment group and model group of elder. And C57BL/6J mice have been divided into two groups as the normal groups in senior and elder. Observe the neurobehavioral manifestation after continuous infusion12weeks by step-down test and step-through test and Morris water maze. When the senior group mice were32weeks and the elders were44weeks, the mice brain tissue were tested by SOD, GSH-Px and MDA. Observe the shape and number of the nerve cells in the hippocampus area by toluidine blue staining. And test the expression of8-hydroxy deoxyguanosine by immunohistochemical method.[Result]1. Step-down test and step-through test result show that the treatment group and model group and normal group in senior have not differences before drug intervention. After continuous infusion12weeks, there is difference between treatment group and model group. The treatment group and normal group still have not differences. But in elder group, treatment group and model group have difference with normal group before intervention. After continuous infusion12weeks, treatment group and normal group have no difference with eath other, but both have difference with model group.2. Morris water mazeresult show that the therapy of tonifying kidney and removing stasis and reducing phlegm could significantly shorten the escape latencyin the place navigation test in the senior and elder treatment group. Compare with the normal group, there is no obvious difference. However, there is a significant difference between treatment group and model group. And the tendency is obvious as time go on. In spatial probe test, the swim distance of the treatment group have been prolonged, the swim time and through time have been enhanced. Compare with the normal group, there is no obvious difference. However, there is a significant difference between treatment group and model group as same as the place navigation test.3. The level of oxidative stress result show that the treatment group and model group and normal group in senior have not differences. But in elder group, the levels of oxidative stress in treatment group and normal group have no difference with eath other, but both have difference with model group.4. The shape and number of the nerve cells and nissl body in the hippocampus area result show that compare with the treatment group and normal group, the express of nissl body in the elder model group significantly decrease by toluidine blue staining. Under the microscope, we can see the nerve cells arrange loose, level of disorder in the elder model group.5. Basic information of inclusion pat ients:the clinical research show that there is no difference between the herbs group and the lifestyle intervention group about age, education degree, weight, diet, sport and history disease.6. TCM syndrome type statistics and score:The majority of patients in this study for the composite card type, and the highest proportion of type is deficiency of the kidney. In terms of syndrome curative effect of traditional Chinese medicine, Chinese medicine group intervention group is better than life s ignificantly.7. Scale assessment:MMSE and MoCA show that all cognitive medicine group after treatment than before treatment significantly improved. Life intervention after the intervention group were improved earlier, but had no statistical difference8. Oxidative stress level:Before the treatment, Chinese medicine group and intervention group life no difference between the oxidative stress level. After treatment, Chinese medicine group of the oxidative stress indicators have improved significantly.[conclusion]1. The conclusion of step-down test and step-through test is ApoE knockout mice have cognitive impairment when they get old. The therapy of tonifying kidney and removing stasis and reducing phlegm can improve neurobehavioral manifestation and cognitive impairment on ApoE knockout mice.2. Morris water maze conclusion is comparing with model group, ability of study and memory in the treatment and normal group mice obviously increase. The therapy of tonifying kidney and removing stasis and reducing phlegm can improve memory and study skill in some extent.3. The level of oxidative stressconclusion is ApoE knockout mice have more oxidative damage when they get old. The method of tonifying kidney and removing stasis and reducing phlegm can improve oxidative damage on ApoE knockout mice. 4. The conclusionof shape and number of the nerve cells and nissl body in the hippocampus area is the therapy of tonifying kidney and removing stasis and reducing phlegm can improve the pathological shape of brain tissue.5. TCM syndrome type statistics and score:Tonifying kidney and removing stasis and reducing phlegm can obviously improve TCM syndrome types in patients with mild cognitive impairment, integral has improved significantly, although lifestyle intervention of MCI TCM syndrome integral has certain improvement, but the effect is not as good as the kidney and phlegm and dispelling stasis method significantly6. Scale assessment:Tonifying kidney and removing stasis and reducing phlegm will greatly improve the score of neuropsychological scale. Lifestyle intervention ability to above a certain degree of improvement, but the effect is not obvious7. Oxidative stress level:Tonifying kidney and removing stasis and reducing phlegm will improve oxidative stress level of MCI. Lifestyle intervention can improve to a certain extent oxidation level.
