In mammalian reproductive cycle,mammary epithelials experience the process of repeated growth,differentiation and degradation.The lactation and the development of mammary gland was regulated by a variety of hormones and cytokines.The three major systemic hormones directly stimulated the development of mammary gland:estrogen,progesterone,and prolactin.These systemic hormone is probably through the tissue regulator,such as FGFs to regulate cell function. All of the FGFs family are the mitogen,affecting the ectoderm,entoderm and the mesenchymal cells.In mammary gland development process,KGFR and its matrix ligand-FGF7 and FGF10 play an important role for mammary epithelial cell proliferation and morphogenesis.The mice were used as experimental animals in this study,through the experiment in vitro and in vivo,systemic invested the dynamic changes in expression and localization of FGF7,FGF10 and its receptor in mammals development,lactation and involution;revealed the relationships between the expression of FGF7,FGF10 and its receptor and the function;clarified the function of FGF7 and FGF10 in mammary gland development,lactation and involution,and the effect of mammogenic hormones on expression of FGF7 and FGF-10 and its receptor in different periods in vitro.The results were as follows:(1) The expression of FGF7 had a few notable high points:virgin 20d,virgin 40d,the early and middle pregnancy,early lactation,and the initial and the late involution.The expression of FGF7 increased to the highest value in involution 4d among these periods.The significantly higher expression points of FGF10 were:virgin 20d and 35d,pregnancy 4d.In lactation 4d,the expression of FGF10 had been relatively high,and the significantly higher expression point was lactation 10d.After entering involution,FGF10 was significant declined in involution 4d and 7d and then increased and maintained a high level expression.The higher expression of KGFR in virgin was 20d,60d;In pregnancy,the KGFR level cutted down slightly first,and increased significantly after pregnancy 10d,then declined slightly after up to pregnancy 16d.In lactation,the expression of KGFR was highest in 4d and then fallen untill involution.The expression of KGFR increased significantly from involution 19d.Partial correlation analysis showed that the expression of FGF7,FGF10 and KGFR were relevant in lactation.(2) FGF7 mainly localized in the matrix of mammary gland ductal epithelial cells and acinar cells.FGF10 showed a similar location pattern as FGF7.KGFR occurred mainly in the mammary gland ductal epithelial cells and acinar ceils,and a weak expression in matrix in the individual time.(3) FGF7 promoted epithelial proliferation and differentiation of mouse mammary.FGF10 was the same as FGF7,and related with fat tissue formation.(4) Except virgin and pregnancy,FGF7 and FGF10 treatment could significantly induced mammary epithelial to produce KGFR.(5) FGF7 and FGF10 can reduce the apoptosis in mammary acinar cells.(6) In mammary glands of mouse,17 beta-estradiol increased the expression of FGF7;progesterone didn't affect the expression of FGF7; In pregnancy and lactation,prolactin upregulated the expression of FGF7 significantly.In mammary gland of mouse,17 beta-estradiol increased expression of FGF10;in virgin mammary gland,progesterone and prolactin reduced the expression of FGF10 significantly;in pregnancy mammary gland,prolactin significantly increased FGF10 expression.When estrogen in the body was relatively high proportion,it would lower KGFR expression;While estrogen in the body was relatively low proportion,it would increase KGFR expression.Low concentration of progesterone increased the expression of KGFR,and high progesterone didn't affect the expression of KGFR.In pregnancy and lactation of mammary gland of mouse,prolactin increased KGFR expression significantly.
