Abstract/Summary:
Classical swine fever (CSF) is a highly contagious disease in pigs caused by the CSF virus. Itwas first discovered in the state of Ohio of America. The disease spread worldwide and its maincharacteristics of are high morbidity and mortality. Bring severe economic losses to the pig farmingworldwide. CSF spread severely in our country and about 39.65%of pigs died of disease died of CSF.The economic losses reach a hundred million RMB. The lapinizated low virulent vaccine wasdeveloped in 1956 in our country and was accepted the CSF safe vaccine using all over the world.Many countries have eliminated the CSF by this vaccine. Although our country have get a bigachievement in prevention of CSF using this vaccine but the CSF are still prevailing in most field ofour country. We did not control prevail of CSF by injecting vaccine in a large area. This lead to doubtthe immuno-efficiency of hog cholera lapinized virus(HCLV).At present, China's main method control classical swine fever is vaccine injection, that artificialactive immunization, As for it is no final conclusion that the difference of domestic CSF immuneeffects with advanced immunization and ordinary routine immunization. And it will take enormousmanpower and resources for farms to take advanced immunization, and there is a possibility of certainanaphylactic response. It is not easy extensive, the immune programme of each hoggery is difference.Currently, new progress of classical swine fever immunization study has been made on the world,But the monitoring for hog cholera antibody is only to detecting the level of IgG. The immunecompetence can not be measured depend on antibody titer of the specific serum IgG only, it includeassociation of IgA, IgM, T lymphocytes and natural killer cells and other cytokines. To enhance therigorous, scientific and comprehensive of the comprehend evaluation of the immune effect ofadvanced immunization for classical swine fever. The means to detect the humoral immunity, cellimmunity and mucosa immunity has been established in the experiment. Routine detecting techniquefor cell immunity such as white blood count and differential, lymphocyte blastogenesis test has beenused, and combine with flow cell technology to detect the level of cell immunization after advancedimmunization at several important aspect, and tracking experiments to observe changes in the numberof the WBC and lymphocytes, lymphocyte proliferation, the dynamic changes of CD4~+ and CD8~+lymphocyte subsets andγ-IFN; and detected dynamic changes rules of IgG, IgM in humor andmucosa with indirect ELISA, then compare with ordinary immunization methods. The results of experiment show that:(1)As the humoral immunity, The levels of antibody in piglets Immuned with days increase withgrowth days in 0-20. In 20-35 days, the antibody level of advanced immunization group is higher thannormal immunization group,as advanced immunization group generated active antibody and maternalantibody of normal immunization group has been counteract, but the antibody level of normalimmunization group is still higher than protection line. At 35~60 days, the antibody level of advancedimmunization group is lower than normal immunization group, but still higher than protection line.After 60 days, the difference of the two groups antibody level is not quiet. The level of IgG is higherthan IgM in detecting.(2)In cellular and molecular level, immune response level of advanced immunization group ishigher than normal immunization group within 20 days; Immune response level of advancedimmunization group is higher than normal immunization group at 30~60 days; the difference of thetwo group is not quiet after 60 days.(3)In the mucosal immune, the mucosal antibody level of advanced immunization group ishigher than normal immunization group within 5 days; the mucosal antibody level of advancedimmunization group is lower than normal immunization group at 30~60 days. the difference of thetwo group is not quiet after 60 days. Tear fluid is most stable and representative in the detectingsimples such as tear fluid, blenna narium, oral cavity fluid, urine and dejecta.(4) In the detecting of humoral immunity, cell immunity and mucosal immunity. The immuneresponse level of advanced immunization group and normal immunization group are all higher thancontrol group. It certify the immune response is specific and generated with swine plague vaccinemotivation. It certify the immune response is specific which generated with swine plague vaccinemotivation. And the dynamic changes rules of the three detecting methods are roughly conformity.In summary, under the circumstances that protect of swine plague vaccine for sow, the differenceof immune response effect between advanced immunity and normal immunity is not quiet. This willprovide evidence and reference not only for illuminate the reaction mechanism of humoral immunity,mucosa immunity and cell immunity reduced by swine plague vaccine, but also for laboratory reseach,immunoprophylaxis and integrate control.
