Mature embryos of 31 wheat genotypes were cultured to compare the efficiency of induction, differentiation and regeneration of mature embryos callus among them. The factors (the genotype, method of induction,concentration of plant growth regulator and so on) affecting the induction, differentiation and regeneration of mature embryos callus were studied. The genotypes, that the efficiency of induction, differentiation and regeneration of mature embryos callus were high, were screened. The mature embryos callus of genotypes screened were transformed by Agrobacterium tumefacienens-mediated (GV3101/pBI121::gus). The factors (A. tumefaciens cell density, inoculation duration, pre-cultivation duration, genotypes of transformed receptor, treatment of negative pressure and so on) influencing the efficiency of transformation mediated by A. tumefaciens were researched. The results showed that1. The genotype, method of induction, concentration of plant growth regulator and so on were important influence factors for the induction, differentiation and regeneration of wheat mature embryos callus. 12 choiceness wheat genotypes (TS021,HB188,SN2618,HB215,Yumai 54,T9817,SN03J102,4072,Yannong 19,HB341,Jinghua 1,T. dicoccum) with the high efficiency of induction, differentiation and regeneration of mature embryos callus were selected out in 31 wheat genotypes via embryo-isolated method (EI) and endosperm-supported method (ES). The optimal method of induction, differentiation and regeneration of mature embryos callus of these genotypes were made sure. The system optimized relatively of induction, differentiation and regeneration of wheat mature embryos callus was established.2. The genotypes of transformed receptor, treatment of negative pressure, pre-cultivation duration, A. tumefaciens cell density and inoculation duration were important influence factors for transformation of wheat mature embryos. The transformed receptor of different genotype had different sensitivity to the same Agrobacterium strain. The treatment of negative pressure in infection could increase the ratio of transformation.
