The Self-Incompability widely exists in plant kingdom and causes an extensive concern in the domains of botany and molecular biology. It facilitates the evolvement of vegetative heredity and extends the variation degree; therefore it has a promising applied prospect in breeding. Some studies indicate that vegetative Self-Incompabilities are determined by S-locus, meanwhile many S-gene fractions and their expression products have been separated. In sporophytic Self- Incompability plant, the S-gene which codes S-Iocus glycoproteins (SLG) in pistil has been obtained, including SLG-2, SLG-22, SLG-29 in Brassica oleracea, and SLG-8, SLG-9, SLG-10 in cabbage. In addition, the S-gene of coding S-receptor kinase (SRK) in pistil is also obtained from the Brassica . SLG and SRK may take part in certain signal conduction in phosphorylation and dephosphorylation, and finally they prevent the growth of the self-pollination. Although the separation of SRK substrate is very important to disclose the mechanism of Self- Incompability signal conduction, but up to date there isn't any achievement made yet.This study completely explores the physiological-biochemical characteristic of protein phosphorylation both in self-pollination and cross-pollination of Self-Incompability Brassica oleracea L., and represents the separation of protein, the substrates of SRK, with pollination and holding self-pollinated pollen back. The results of the study are significant and will be discussed from the following five related aspects.1. The relation between the pollen germination of Brassica oleracea L. Self-Incompability and protein phosphorylationThe impact on pollen germination and pollen tube growth is discussed respectively through adding orthophosphoric acid, ATP, CaCl2 and quercetin into the extraction of style. Meanwhile, the function of protein kinase at inhibiting the self-pollination germination and the relation between thepollen germination and protein phosphorylation are also analyzed.The result indicates that the germination ratio of self-pollination and the growth of pollen tube decrease obviously through the treatments of ATP, CaCl2 and ATP + Na3PO4, but the germination and growth of cross-pollination pollen increase. Quercetin has greatly promoted the germination ratio and the comparative length of pollen tube in self-pollination, but a few influence on cross-pollination. So we can infer that SPK kinase is essential to Self- Incompability.2. The change of physiological-biochemical characteristic in style caused by Self- Incompability Brassica oleracea L. with protein kinase inhibitorThis section studies the dynamic variation of enzyme protection system, which is the style of Self- Incompability Brassica oleracea L, after pollination in both self-pollination and cross-pollination. The contents of SOD, POD and CAT enzymes, which strengthen the abilities of the enzyme protection system, rise up after pollination in styles. The contents of three enzymes in cross-pollination rise higher than that in self-pollination. With a view to the constitutes of SOD, POD and CAT enzymes, we find out POD content exceed the SOD and CAT contents comparatively inside the style of Brassica oleracea L., and these phenomena occur more obviously in self-pollination.The dynamic variations of Ca2+ and K+ contents inside the style of Brassica oleracea L. during pollination process are observed by using ashing-technique and atomic absorption method. The result reveals whether SI Brassica oleracea L. of self- pollination or cross- pollination, the K+ content is in a descending trend after pollination. K+ content descends quicker after 0-1 h pollination in self-pollination than that in cross-pollination. After 3-6 h pollination, there is an extremely significant difference of K+ content between self-pollination and cross-pollinationOn the other hand, whether self- pollination or cross- pollination, the Ca2+ content will reduce rapidly afterO-lh pollination, and gradually reduce along the time after pollination.
