As idea-point and base for main proposition in the study for problems ofsafety on water and harm for zoology, especially pollution actuality on waterresource and water disposal technic in worldwide, operating situation onpurificant for water. The paper engaged study with Jilin university innovationfoundation of P.R.China .Taking one with another for domestic study on microbe purificant, Thereports in all-around and screening on thalli are more than reports on experimentstudy and appraisal work, composing of culture medium and action mechanism,study on product extraction. The flocculant function was single only to wipe offsuspender and color in water, but not take away heavy metal ion and organicpollutant effectively. in particular, it is non report on system study deeper whatpurificant was made of compound microbe.1.The object and main content on studyThe statement of the paper turns toward system's study deeper on high effectcompound purificant which flocculate and wipe off organic pollutant withinclusion of adsorbing heavy metal ion (for instance Cd,Cr,As,Pb etc) in water.Main content on study: screening compound bacteria with function on higheffect flocculation and wiping off organic pollutant, adsorption to heavy metalion;ensuring ferment condition and ferment parameter for compound bacteria;engaging study on characteristic of physiology -biochemistry and extremeenvironment to compound bacteria;component and safety on purificant;clearing effect on product purificant of compound bacteria;cataloging on geneto composing bacteria and the more to gene level appraising and establishment ofsystem growth tree;The results of experimentation for various disposal effect onwater quality will provide scientific gist to mid-test. The study on succeed ofpurificant will take complementarity and sublimationon theory which fill blank indomestic theory on compound microbe purificant. The extension and operationabout it will act on citizen's water and old industry base in northeast part andelsewise on environment which it will show picture of benefit on environmentand society and economy.2. The results of main experimentationResult No.1It is made up of 3 kinds of bacteria including 2 kinds of actionomyces and 1kind of microzyme where adopted from polluted water irrigative area in a city. 9kinds of haploid clearing bacteria filtrated after flocculating and zymolysis andculture where cooperated with central lab of environment and resource college inJilin university. Then 9 kinds of high effect compound clearing bacteria filtratedafter combination of C92and C93. The farther work of seeing about compositiveproportion of compound bacteria compared with produced clearing preparation ofhaploid flocculated bacteria and applied on first step, at last objective compoundbacteria was confirmed.Result No.2To engage study of culture condition on the compound bacteria, the resultis that the best components of zymolysis culture medium are dextrose, 2.0g;K2HPO4·3H2O 5.0g;KH2PO4 1.0g MgSO4·7H2O 0.2g;Distilled water 1L,pH=7. Culture condition in Outside of Complex bacterium: temperature was20°-30°C while pressure of destroying bacterium was 0.06Mpa and rotationalspeed was 100-140r/min, capability of the liquor is 150mL.As the culture condition above to study change of pH and basic elementincluding dextrose and content of ammonium sulfate, activity of compoundmicrobe flocculant, change of output in different term. The results in wholeupgrowth Cyc showed that pH was changeless , about 7;source ofcarbon-nitrogen was obviouse that content of carbon source consumed morequick and its rate was biggish, change rate on content of nitrogen sourceassorted with first one;the top extent of flocculent activity is wide and itsmax.was 97.35% as well as its output goes to max. 3.02g/L, relevant invert ratewas 68.18%. The results of test provided base to study flocculation effect and itsmechanism more.Result No.3The results of study on character of physiology and biochemistry forcompound bacteria and upgrowth circs of extreme environment show that productbacteria of compound microbe purificant was changeless on character ofphysiology and biochemistry after combination,only some roles more stronger.In 40~160 MPa high pressure, form and quantity, dead rate on productbacteria of compound microbe flocculant existed influence obvious as well as onits activity on microbe flocculation and flocculation activity on product purificantof haploid bacteria existed pressure's threshold point, but some part to characterof physiology and biochemistry on bacteria,furthermore,haploidbacteria's sort ofcompound microbe flocculant under pressure of activity of flocculation existedthreshold obviouse, but its product bacteria had no rule. It provided theoryparameter to culture and application, new leading change way on productbacteria of compound microbe flocculant under high pressure's environmentthrough the study of the paper. It is well to upgrowth environment in high metal'sliquor and able to wipe off heavy metal ion on upgrowth process.Result No.4The result of analysis in qualitative and quantitative way on glucide andprotein in tradition to purificant using infrared spectrum issued that the effectivecomponent of compound microbe purificant was amylose matter within part ofalbumen, total content of glucide was 82.3% and protein was 7.8%, itsstructure was linearity. otherwise it was differential on result between productmatter of haploid bacteria showed and product matter of compound bacteria.Flocculation effect of Purificant for water was well by heat of 40-100℃ in 30mins, that is hot stability of it is well. The other result of experimentation onlittle white rat's toxicology showed that they were all right for psychosis and nonexceptional reaction about posture and eating or action, they like walk properlyunless eating and non death Ld50>1g/kg. The purificant for water by securityexperimentation is safe-innocuity to apply widely on drinking water source andtiny polluted water or waste water etc.Result No.5The result of study on effect of product pyrificant of conpound bacteria forwater and purge mechanism is following:Purificant took kaoline as flocculation object to research flocculation effectand flocculation mechanism and flocculation kinetics, their results issued that:(1). Study on flocculation effect: best aid-coagulator of purificant was CaCl2and optimal addition was kaoline ( 5g/L 1L) / (10% CaCl2 liquor) 0.75mL;best pH was 6-7;best addition of purificant was 8.0 mL;temperature 4-60℃;mix –coagulator rotational speed was in 50-175r/min.(2) Study on flocculation mechanism: comparison between observation underoptical microscope and incompact structure of kaoline before flocculation underaction of purificant and Ca2+, it formed a agglomerate from inside to outside tointerplay on kaoline's big granule and big granule, big one and small one, smallone and small one. This kind of conection by high temperature mix never beinfluenced. It issued that form of agglomerate is very close and solid to providedgood result for flocculant application.Analysis on scanning electronic lens was same to optical microscope,Absorbability scale and position on a collection of illustrate plates atlas of kaolinewas change under action between purificant and Ca2+.As the purificant was able to flocculated only when Ca2+ existed, so theflocculation mechanism is almost to alginate's.(3) study on flocculation kinetics: the effect was to added Ca2+ and purificantat rotate speed 100rpm in experimentation. It means that most offlocculation took sedimentation by circumgyrating mode in shearing force whenpurificant and Ca2+ and laoline were agitated, few of it by gravity, so kineticstheoty on product purificant of compound microbe flocculated kaoline is main onoverfall theory.2.study on effect of which purificant adsorbs 4 kind s of heavy metal ion,adsorption mechanism and adsorption kinetics(1)influence of adsorption effect:best rotate speed:adsorption Cd( Ⅱ ):70r/min,As( Ⅲ) :110r/min,Pb( Ⅱ) :120r/min,Cr( Ⅵ ):50r/min. best point oftemperature:Cd(Ⅱ):40℃,As(Ⅲ):20℃,Pb(Ⅱ):20℃。Cr(Ⅵ):10℃;best time of purifican textracted by zymolysis liquor:Cd(Ⅱ):24h,to adsorb other3 kinds of heavymetal ion:60h;best point on pH:Cd(Ⅲ):pH=1, As(Ⅲ):pH=4,Pb(Ⅱ):pH=6,C(rⅥ):pH=5,;Best additionof purificant:Cd(Ⅱ)maximaladsorption point was 29.44mg/g while addition of purificant was 1mL;As(Ⅲ)maximal adsorption point was 14.79mg/L while addition of purificant was3.5mL;3.5mL.Pb(Ⅱ)maximal adsorption point was 171.63mg/g while addition ofpurificant was 0.5mL;Cr(Ⅵ)maximal adsorption point was 7.54mg/g whileaddition of purificant was 1mL 3mL.The result on what coexistence ioninfluenced adsorption issued that it had appeared to positive action or minusaction when 2 kinds of heavy metal ion was mixed, and it accelerated on thewhole what purificant adsorb heavy metal ion when more than 3 kinds of heavymetal were mixed(2)Study on adsorption mechanism;the result of infrared testing issued thata collection of illustrative plates atlas of infrared is typical collection ofillustrative plates atlas of polysaccharid, component of the purificant hashydroxy, respectively as —COO-and COOH . intension was changed afterrelevant chart apex make point moved, becoming action point of bothpurificant's function agglomerate and 4 kinds of relevant metal ion. It appreared many globularity samll granule after Cr wasadsorbed, it can be conjectured to big molecule object formed with chromium onsurface of purificant, these big molecules boil with Cr, and then formedorganic crystal Cr, take change of space on surface of purificant's granule.(3)Study on adsorption kinetics: purificant adsorbed 4 kinds of heavy metalion by passiveness adsorption mode.(4)Analysis on adsorption isotherm: best model o Cd's was Temkin, bestmodel of As's was Frendlich, best model of Cr's was Langmuir andTemkinResult No.6It engaged geneticsstudy on activity point of clearing effect on productpurificant of compound bacteria, the result issued that plasmid was action geneof controlling clearing effet.It engaged to type gene for compound bacteria by RAPD technology, theresult is that 3 kinds of bacteria existed congenetic character, but differentialcharacter was distinct. SrDNA appraisal and establishment of system growth'stree gave 3 kinds of bacteria appraisal on gene levelResult No.7As clearing object to kaoline suspended liquor, comparison with traditionalpurificants, for instance PAM, Al2(SO4)3,FeCl3,Its result issued that purificanteffect of compound bacteria was excellent and non-toxicity.To engaged series experimentation on application area of the purificant, itsresult issued that the purificant could applied to familiar polluted water, forinstance, chromium balck T water and lysine water, amylum waste water andliving waste water, heavy metal waste water .etc. Because of compound microbepurificant is non-toxicity, so it is very condign to apply on tiny waste water anddrinking water.3.Innovation point on question for discussion(1)To engaged deeper discussion on flocculation bacteria's physiology andbiochemistry characteristic, growth Cyc and sort position. To take the lead indoing study on 3 kinds of jointly made compound microbe purificant.(2)It proved that compound microbe purificant has integrative clearing effectby study on clearing effect ( for instance on flocculation suspended object,wiping off heavy metal ion and organic waste object.(3)It proved that compound mmmicrobe purificant belong to friendlyenvironment material on safe ecotype by experimentation on analysis ofpurificant component and toxicology.
