Based on the recombinant pcDNAKCpG constructed, the heredity stability, survival ability and its persistence, distribution and the potential integration into host genomic DNA were studied. The main conclusions can be summarized as follows:1. After 30 passages, the copies of the recombinant pcDNAKCpG and the sequence of CpG ODN didn't change, and the mutation and loss of the kana resistance gene haven't been found also, so the pcDNAKCpG has good heredity stability.2. After the pcDNAKCpG was injected into the chickens and pigs, the recombinant plasmid was mainly found in the whole blood and the injected muscle, and the persistence was no more than 16days and 8 days, respectively.3. PCR detection showed that no pcDNAKCpG could be detected in the environment bacteria and in host genome of all the tissues.4. After stored at -70°C, -20°C and 4°C for two months, the livingness of recombinant E.coli containing pcDNAKCpG didn't change.5. The recombinant plasmid has an enhanced effect to immune response.6. After pcDNAKCpG was injected into the chickens and pigs, no abnormal behavior could be observed in above animals, therefore, the plasmid has a good biosafety.
