Haemorrhagic fever with renal syndrome is a severe infectious disease and still can't be cured effectively. Several mouse hybridoma cell lines, which secrete McAb specified for HFRS virus, have been made since 1980s, and have been applied in clinical diagnosis extensively. Some of them with high activities of hemagglutinative inhibition and neutrolization have showed protective activities while the animals were challenged with HFRS virus. It indicated that administration of protective McAb may be helpful to the treatment of HFRS. However, a major factor limiting the application of the mouse McAb to human is the development of an anti-antibody response in recipient. In an attempt to overcome the immunogenicity, the mouse McAb should be 'humanized' or fully human McAb should be made.In this study, the VL and VH genes of mouse McAb 1A8 against HFRS virus were amplified, cloned, sequenced and ScFv were expressed on phage...
