It is known that DAG/PKC signaling pathway is very important in signal regulating network in cells. It involves in the growth, proliferation, division and carcinogenisis of cells, and distributes abroadly in all tissues. In this study we mainly studied the effects of Le~y oligosaccharides and PEMT2 overexpression on the DAG/PKC signaling pathway.Embryo implantation is the key step of reproduction, which includes the synchronized developmental process of embryo and endometrium. Lewis oligosaccharides are fucosylated lacto-series oligosaccharides including H1, H2, Le~a, Le~b, Le~x, sLe~x and Le~y et al. Recent study showed that Lewis oligosaccharides play an important role in the embryo implantation, especially H1, Le~x, sLe~x and Le~y are closely related to the successful implantation of embryo. The implantation rate of mouse embryo was obviously reduced by incubating with anti-Le~y monoclonal antibodies in co-culture system in vitro or injecting the antibody into uterine lumen in vivo, but the regulation mechanism was still unclear.The synthesis of nucleic acid and protein is defined by the templates those determine the orders of nucleotides and amimo acids, but when it comes to carbohydrate chain, the template is deficient and a series of ordered highly specific glycosyltransferases function. The synthesis of fucosylated oligosaccharides is decided by the fucosyltransferases, so the expression of fucosyltransferases might be the key point of the synthesis of oligosaccharide antigens and the regulation of oligosaccharide antigen synthesis. At present, there are 11 fucosyltransferases (FUTs) have been identified. In mice, FUT1, 2, 4, 7, 8, 9 are functionally expressed, the others are pseudogenes. In these fucosyltransferases, FUT1 and FUT4 take part in the synthesis of Le~y oligosaccharide, FUT7 takes part in the synthesis of sLe~x oligosaccharide, while FUT9 is the key enzyme of the synthesis of Le~x...
