Expression of c —fos gene in functional networks of the central nervous system of the rat in vivo, neurons in vitro and the mechanism was studied by using immunocytochemystry, microin-jection, retrograde or transganglionic labelling of CB —HRP, and other methods. In order to further research, a nonradioactive fos probe was prepared and used for detecting fos mRNA by in situ hybridization. The present work consists of eight parts. A :otal number of 112 Wistar rats and 78 dishes of cultured cerebral and hippocampal neurons from newborn rats were used.I . The temporal and spatial distribution of Fos protein in pain sensory —nociceptive and responsive system,after a limited noxious stimulus (a puncture) applied to the left tongue tip of conscious rats, was studied. Expression of c —fos was induced in ganglion of trigeminal nerve and widespread brain regions including where once known or not as nociceptive, regulative and responsive regions (e. g. ,caudal part of the trigeminal nu. , periaqueductal gray, et al). The nuclei and regions that are related to movement, vision and sense of hearing were alsc involved. Special part of ependyma cells were activated to express c —fos more lately. But the ventral postero-lateral thalamic nucleus had no labelled cells. The distribution of Fos — IR labelled cells in the medulla oblongata was in accordence with CB—HRP labelled axon terminals from transganglionic transport except the postrema area. Most of the regions that presented dense Fos —IR labelling were the regions presenting dense CGRP—IR and/or SP—IR terminal.I.
