Hepatitis B virus(HBV) is a DNA virus. The HBV genome is a single 3,200bp, partially double-stranded, relaxed-circular DNA, which encodes polymerase, X protein, core antigene, and suface antigens. It replicates within infected liver cells. Effective and safe vaccination for hepatitis B is available for more than 20 years, nonetheless, hepatitis B caused by HBV is still a serious fatal disease largely because therpapeutic intervention options for chronically infected HBV carriers are rare and usually of limitied success. It is estimated that 3.5 billions people worldwide are infected with HBV each year and especially in china about 50-70% persons has been infected with HBV and 8-12% persons , that is more than 1 billion person, are carrier of hepatitis B surface antigen. More than 1 million persons will die of sequeal related to HBV infection. At the current, there are 3 available drugs USA FDA-approved medications for the treatment of hepatitis B: recombinant interferon, nucleoside or nucleotide analogs including lamivudine and adefovir dipivoxil. Interferon and adefovir dipivoxil inhibit the HBV reverse transcriptase and DNA polymerase. However, even with different drug combinations there is limited effectiveness not suited to achieve complete cure from the virus. This is because they do not promote HBV eradication from the infected host, thus resulting in relapse and recurrence of viremia after cession of treatment.
