By now, there are no efficient medicine or medical measures to treat rotavirus-induced diarrhea in infants. Therefore, rotavirus vaccine research is still the focus in this field. Rotavirus vaccine research involved attenuated live vaccines, subunit vaccines, genetic recombinant vaccines, DNA vaccines and viral vector vaccines. The purpose of these different kinds of vaccines is to elicit rotavirus specific immunity. As a gastrointestinal infective virus, local secreted antibodies play special and important roles to defend rotavirus infection. This paper studied VP4 gene modification, VP4 expression in prokaryotic and eukaryotic systems, and evaluated the immunogenicity of recombinant VP4 in mice and guinea pigse.VP4, the outer capsid protein and hamegglutinin of rotavirus, plays important roles in virus absorption and virulence. We choose rotavirus SA11 strain VP4 gene as the target gene. The length of SA11 rotavirus VP4 gene is 2363 base pairs (bp). Mature VP4 comprises 776 amino acids (aa), and the molecular weight is about 87kD. To improve the immunogenicity of VP4, we modified VP4 gene in two ways.
