Firstly we research the epigenetics regulation of 14-3-3 σ in animal cells, next we research Differential expression and epigenotics model of 14-3-3σ induced by shRNA. Forestall we found that the expression of 14-3-3 σ owed to the methylation model in its CpG island. We also found that the expression of 14-3-3 σ was owned to its histone modification in its promoter .These works may suggest that 14-3-3 σ is regulated by epigenetic factors. We introduced shRNA expression plasmids stably into human cells, we detected significantly DNA de novo methylation within the 14-3-3 σ promoter , the deacetylation of histones happened in the promoter of 14-3-3 a, the expression level of 14-3-3 σ mRNA was significantly lower than in wild-type human cells (down~50%). Our study will give us a better understanding of the mechanism of RdDM, would greatly help in the development of therapy of tumor. |
