Multicellular animals often need to get rid of cells that are in excess, in the way , or potentially dangerous. To this end, they use an active dedicated molecular program. Cell deaths during embryonic development are essential for successful organogenesis and crafting of complex multicellular tissues. The evolutionary advent of differentiated cell types may have necessitated controlling death as well as division in order to keep neighboring cells interdependent and insure the proper balance of each cell lineage. Apoptosis also operates in adult organisms to maintain normal cellular homeostasis. This is especially critical in long lived mammals that must integrate multiple physiological as well as pathological death signals, which for example includes regulating the response to infectious agents. Gain and loss of function models of genes in the core apoptotic pathway indicated that the violation of cellular homeostasis can be a primary pathogenic event that results in diseases. Evidence indicated that insufficient apoptosis can manifest as cancer or autoimmunity.According to if there is cysteine protease involved in apoptosis pathway, which can be divided into caspase dependent or casapse independent apoptosis; Depending on the sequence of cell death signaling transduction , apoptotic pathway can also be divided into upstream or downstream pathway. In general, signal derived from mitochondria will be considered as downstream apoptotic signal, such as release of cytochrome C from mitochondria mediated caspase cascade reaction. The signal mediates mitochondria dysfunction and releasing death promoting molecular will be thought as upstream apoptotic signal. The downstream apoptotic pathway is quiet clear at present, but the upstream apoptotic pathway is still vague.In order to investigate apoptosis regulation and search apoptosis related genes, we designed a procedure for identifying genes required for STS and UV induced cell killing, using retroviral random insertion could generate genomic mutations.
