Using thorough genetic experiment, genetic effect was studied on T.spelta var.duh. 1BS chromosome segments; The fertile change mechanism and range applied and genetic specific property of thermo-sensitive were conducted through multi-locus, multi-locus sowing at different time and redeveloped tillers of the cut tillers at different time. And The RAPD techniques were used for molecular marker of wheat thermo-sensitive male sterile gene. Analysis on the findings of these experiments shows that: 1. By applying the technology of Chinese invention patent 00105488.0, Genetic effect were studied on breeding Non-1B/1R K type male sterile wheat line by means of chromosome transfer. The result indicated that T.spelta var.duh. 1BS chromosome make Non 1B/1R K type male sterile wheat line have better fertility-restoring feature and less haploid. There were better genetic effects on almost all agronomic characters except for the plant height.2. To establish the two-lines hybrid wheat seeds production system, which is easy-use and low-cost, and to breed the thermo-sensitive male sterile wheat lines that could be used in wide-range areas, experiments on the thermo-sensitivity of the thermo-sensitive male-sterile wheat line A3314, which was bred Correlation Analysis on the selfed seeds rates of 10 different sowing dates and of redeveloped tillers of cut tillers at 8 different dates with the corresponding air temperature, air humidity and day-length during the booting to flowering stage in Yangling shows that: the male fertility of A3314 is thermo-sensitive;The sensitive stage of fertility exchange is in the period of 8 days around booting stage, i.e. from miosis to mononucleate stage;the male fertility of A3314 will be male-fertile while the temperature is over 18.3℃ during the sensitive stage;there is no significant relationship between the male fertility exchange of A3314 and day-length; on the conditions of temperatures over 18℃, there is positive correlation between the selfed seeds rates of A3314 and the humidity while the humidity range in 40-80%. Based on the analysis of the thermo-sensitive characters of A3314.3. the thermo-sensitive male-sterile wheat line A3314, which was bred by applying the technology of Chinese invention patent 00105488.0, were conducted through multi-locus, multi-locus sowing at different time. Analysis on the findings of these experiments shows that: the male fertility of A3314 will be stably male-sterile while sowing at the time suitable for wheat growing in Huang-Huai (the valleys of Huanghe river and Huaihe river) winter wheat production area, Yunnan-Guizhou winter wheat production area, Northwest spring wheat production area and Northeast spring wheat production area, and the secure sowing date to guarantee A3314 male-sterile is around 60 days during the local wheat sowing dates; on the other hand, the male fertility of A3314 will be male-fertile and the selfed seeds rates will be over 60% while sowing in spring or summer in Huang-Huai winter wheat production area and Yunnan-Guizhou winter wheat production area. Based on the analysis of the thermo-sensitive characters of A3314, A3314 will be securely applied in hybrid seed production in most wheat production areas of China.4. Genetic Studying of Thermo-sensitive Male Sterile Gene on Thermo-sensitive Male Sterile Line A3314 by fertility reaction of F2 and BC1F1 population of A3314/Silverstar and K3314A/A3314 under fertility condition. It showed that thermo-sensitive male fertility might be control mainly by two pairs of segregate excessive genes excide for cytoplasm genes, especially rfv1sp driving from T.spelta var.duh. 1BS chromosome and main male sterile gene of K3314 were a pairs of allies, and having thermo-sensitive character.5. Optimization of the RAPD reaction system of wheat. It showed that this system was 25ul total volume contained 50ng template DNA ,2mmol/l Mg2+,0.2mmol/l dNTPS, 0.2umol/l random primer, 1U Tag ploymerase. After pre-denaturing at 94℃ for 5min, under the condition of denaturing at 94℃ for 45 seco...
