Dealing with wild tomato and Nr mutant tomato as materials, This paper studied the ethylene production, expression changes in ethylene synthesis critical enzyme gene, ACS gene family, and nuclear transcription factor gene, under different stress conditions. More detail as follows:1 .The ethylene production of wild tomato germinating seed treated with NaCl and PEG was studied. The experiment showed that salt and osmotic stress increased ethylene production. In addition, the triple response strengthened. The results suggested ethylene was a signal factor that induced tomato germinating seed adaptive response to stress conditions.2.The wild tomato and Nr mutant tomato fruits discs were treated with wound, 0.8 mol/ L NaCl and 0.04 mol/ L PEG. The similar trends in ethylene production between the two types was observed. This indicated that ethylene production under stress conditions could achieved without NR protein.3.The wild tomato germinating and fruit were treat with NaCl and PEG to detect expression changes of LE-ACS gene family. There was changing of LE-ACS1A, LE-ACS1B, LE-ACS4, LE-ACS6 gene expression in transcription level with the growth of the germinating and stress condtion. So the expression of LE-ACS gene family was regulated by stress factor as well as growth factor.4.The wild tomato and Nr mutant fruit discs treated with wound, NaCl and PEG to detect expression changes of LE-ACS gene family. The experiment showed that during the ethylene production under stress conditions, the LE-ACS2 gene expression maintained at a high level, different from system I ethylene production transition to system II. This suggested that stress conditions changed the regulatory mechanism of LE-ACS2.5.The effects of NaCl and PEG in wild tomato germinating seed on the expression of nuclear transcription factors genes, including Le-EILs, LeERFs, was studied. The results showed that all the gene responded to the growth factors and stress factors, suggesting the they might play their roles in at least two signaling pathways.6.The effects of NaCl and PEG in wild tomato and Nr mutant fruit discs on the expression of nuclear transcription factors genes, including Le-EILs, LeERFs, was studied. With respect to wild tomato, Le-ERF2 expressed higher under salt and osmotic stress than under wound stress. And there was a significant difference in expression trends. But Le-ERF2 showed a consistency in the expression trends and level under different stress conditions. These suggested NR protein didn't play a role in Le-ERF2 expression in salt and osmotic condition.
