| Purpose:To investigate the prevention and treatment effect of expectorant and stasis combination in the treatment of rats with atrial fibrillation model,and to study the mechanism of action from the perspective of the prescription’s response to inflammation,which was used to guide clinical practice.Material and method:Sixty male rats with healthy SD were selected,10 in the control group and 50 in the model group were randomized,and the model group was made by Ach(66ug/ml)-CaC12(10mg/ml)mixture for 7 consecutive days by rat tail vein injection to make a rat model of atrial fibrillation.The rats who successfully modeled were randomly divided into 5 groups,including model group,expectorant and stasis high-dose group,expectorant and stasis medium dose group,expectorant and stasis low-dose group and verapamil group,and gastric gavage for 2 weeks,during which tail vein injection was still maintained.After 2 weeks,rats were anesthetized with 10%chloral hydrate,and the limb leadⅡ ECG was recorded using the PowerLab 15T data acquisition and analysis system to observe the duration of atrial fibrillation in each group.Under transmission electron microscopy,the changes of myocardial ultrastructure in each group were observed.The contents of IL-18 and IL-1β in the serum of rats were detected by ELISA method.The Western Blot method detected the expression of ALRP3,Capase-1,ASC,IL-18 and IL-1β proteins in rat atrial muscle.Results:1.Comparison of the results of expectorant and stasis removal of traditional Chinese medicine compound on the duration of atrial fibrillation model ratsAfter 2 weeks of inoculation,the duration of atrial fibrillation in rats in the model group was significantly prolonged compared with the blank group(P<0.05).Compared with the model group,the duration of atrial fibrillation in rats in each treatment group was significantly shortened(P<0.05).2.Effect of expectorant and stasis removal of traditional Chinese medicine compound on myocardial ultrastructure in rats with atrial fibrillation modelThe muscle segments of rat atrial myocytes in the control group were complete and clear,the muscle filaments were arranged neatly and tightly,the mitochondrial structure was normal,and the myofilaments of cardiomyocytes in the model group were disordered or even broken,and the mitochondrial morphology was different and some vacuole-like changes appeared.After 2 weeks of intervention,the changes in the structure of the atrial muscles in the animals in the treatment group were reduced.3.Comparison of IL-18 and IL-1β contents in serum of rats with expectorant and stasisCompared with the blank group,the serum content of IL-18 and IL-1β in the model group was significantly increased(P<0.01).Compared with the model group,the serum contents of IL-18 and IL-1β in the high and medium dose group of expectorant and stasis removal were significantly reduced(P<0.01).Compared with the verapamil group,the serum content of IL-18 and IL-1β decreased in the expectorant and medium-dose group(P<0.05).4.The combination of expectorant and stasis was used to affect the expression of atrial muscle NLRP3,Capase-1,ASC,IL-18 and IL-1β proteins in atrial fibrillation model ratsCompared with the blank group,the expression of ALRP3,Capase-1,ASC,IL-18 and IL-1β proteins in the model group was significantly increased(P<0.01).Compared with the model group,the high,medium and low dose groups and the verapamil group could significantly downregulate ASC and IL-18 protein(P<0.01).The high-dose expectorant group and verapamil group could reduce the expression of IL-1β protein(P<0.05);The expression of NLRP3 protein decreased significantly in the high,medium and low dose groups of expectorant and stasis(P<0.01),while the expression of NLRP3 could be downregulated in the verapamil group(P<0.05).The expression of Capase-1 protein was significantly reduced in the expectorant and high-dose group and verapamil group(P<0.01).Conclusion:1.Expectorant and stasis Chinese medicine compound can effectively intervene in atrial fibrillation induced by Ach-CaC12 and shorten the duration of atrial fibrillation;The combination of expectorant and stasis Chinese medicine can improve the changes of myocardial ultrastructure in rats,inhibit myocardial fibrosis,and reverse structural reconstruction.2.The combination of expectorant and stasis Chinese medicine significantly reduced the content of serum IL-18 and IL-1β in rats,inhibited the occurrence and development of inflammation,and delayed the emergence of complications;The combination of expectorant and stasis Chinese medicine can reduce the expression levels of NLRP3,Capase-1,ASC,IL-18 and IL-1β proteins,inhibit the activation of NLRP3 inflammasomes,antagonize myocardial fibrosis,improve myocardial remodeling,and delay the occurrence and development of atrial fibrillation.The combination of traditional Chinese medicine can effectively intervene in the sensitivity of atrial fibrillation induced by Ach-CaCl2 and shorten the duration of atrial fibrillation. |
