| Objective:To discuss the correlation between inflammasome NLRP3 and atrial fibrillation by detecting the expression level of pheripheral blood inflammasome NLRP3 and other inflammatory factors in atrial fibrillation patients.Method:Sixty atrial fibrillation patients were enrolled in the study.Patients who fulfilled the eligibility criteria were assigned to paroxismal atrial fibrillation group (PAF)(n=30) and non-paroxismal atrial fibrillation group (nPAF)(n=30), and at the same time the healthy volunteers were selected as the control group (n=26).Clinical data were collected and observed include:gender,age,history of present illness,past medical history,personal history,family history,and the positive signs of vital signs and physical examination.Venous blood was drawn in the morning and was centrifuged to obtain peripheral blood mononuclear cells (PBMCs) and serum.flow cytometry was used for detected NLRP3 expression levels in peripheral blood mononuclear cells (PBMCs) and ELISA was used for detected the concentration of IL-1β,IL-6,CRP,NT-proBNP in blood plasma. Color Doppler Echocardiography was used for measure the diameter of left atria, the diameter of left ventricular end-diastolic,interventricular septum thickness,left ventricular posterior wall thickness and left ventricular ejection fraction respectively by professional of ultrasound specialist. SPSS16.0 statistical software were used for analysis,P<0.05 indicates a statistically significant difference.Result:1.Both PAF group and nPAF group of patient’s NLRP3 level in peripheral blood mononuclear cells (PBMCs) were higher than those in healthy volunteer control group (P<0.05),but both PAF group and nPAF group of patient’s NLRP3 level in peripheral blood mononuclear cells (PBMCs) were no statistical significant (P> 0.05).2.Both PAF group and nPAF group of patient’s IL-1β,IL-6,CRP levels in blood plasma were higher than those in healthy volunteer control group(P<0.05),but both PAF group and nPAF group of patient’s IL-1β,IL-6,CRP levels in blood plasma were no statistical significant (P >0.05).Both PAF group and nPAF group of patient’s NT-proBNP level in blood plasma were higher than those in healthy volunteer control group (P<0.05),but both PAF group and nPAF group of patient’s NT-proBNP level in blood plasma were statistical significant (P<0.05).3.Both atrial fibrillation group (PAF,nPAF) patient’s NLRP3 level in peripheral blood mononuclear cells (PBMCs) was positively correlated with left atrial diameter (R=0.579,P<0.05), and was negatively correlated with left ventricular ejection fraction (R=-0.490,P <0.05).Conclusion:1.Both atrial fibrillation group (PAF,nPAF) patient’s NLRP3 expression level in peripheral blood mononuclear cells (PBMCs) were higer than those in healthy volunteer control group,this suggested that NLRP3 inflammasome was closely related to atria fibrillation.And the NLRP3 inflammasome may have positive clinical value in the intervention process of the development and maintaining of atrial fibrillation,and may offer more effective therapeutic strategies for the treatment of atrial fibrillation.2.Both PAF group and nPAF group of patient’s blood serum inflammasome NLRP3 downstream IL-1β and other inflammatory factors like IL-6,CRP levels were higher than those in healthy volunteer control group, this further indicating that atrial fibrillation and inflammation response are closely related.3. The NLRP3 expression levels in both AF group associated with LAD and LVEF,it may be through inflammasome NLRP3’s related inflammatory factors downstream produce inflammatory response,leading to cardiac structure or electrical remodeled,and induce to development and maintenance of atrial fibrillation. |
