Objective:To investigate the effects of analgesic Zhitong Jiangu Decoction drug-containing serum on the expression of IKKβ,NF-κB p65 and TNF-α in synovial fibroblasts,and to clarify whether analgesic Zhitong Jiangu Decoction regulates the NF-κB signaling pathway of KOA synovial fibroblasts,which provides theoretical support and scientific basis for the clinical application of pain relief Zhitong Jiangu Decoction in the treatment of KOA.Methods: After the successful preparation of rabbit KOA model,the synovial tissue in the rabbit knee joint was isolated,synovial fibroblasts were extracted and cultured,and the third-generation cells were taken for experiments,the experiment was divided into 7 groups,the blank control group did not add serum,the normal serum group and the analgesic Zhitong Jiangu Decoction drug-containing serum group were set to 5%,10% and 20% volume fraction concentrations,respectively,and the concentrations of IKKβ,NF-κB p65 and TNF-α in synovial fibroblasts were detected after the culture time of each group was 48 and 72 h.Results: Normal rabbit serum and Zhitong Jiangu Decoction drug-containing rabbit serum can reduce the expression of IKKβ,NF-κB p65 and TNF-α in rabbit KOA synovial fibroblast cells,and its anti-pain and Zhitong Jiangu Decoction drug-containing rabbit serum can play a better inhibitory effect.In the analgesic Zhitong Jiangu Decoction serum,the analgesic Zhitong Jiangu Decoction drug-containing serum with a volume fraction of 20% had a more obvious inhibitory effect on the expression of IKKβ by rabbit KOA synovial fibroblasts than the drug-containing serum with 5% and 10%.Compared with 5% and 10% drug-containing serum,the drug-containing serum of analgesic Zhitong Jiangu Decoction with a volume fraction was more obvious in the inhibition of NF-κB p65 in rabbit KOA synovial fibroblasts.Compared with 5%and 10% drug-containing serum,the drug-containing serum of analgesic Zhitong Jiangu Decoction with a volume fraction of 20% had a clearer inhibitory effect on the expression of TNF-α in rabbit KOA synovial fibroblasts.Conclusion: Zhitong Jiangu Decoction can reduce the levels of IKKβ,NF-κB p65 and TNF-α in KOA synovial fibroblasts.
