Objective: Through the retrieval of China Knowledge Network(CNKI)and Wanfang Journal database,the literature about the treatment of type 2 diabetes mellitus by traditional Chinese medicine from liver was obtained,and through data mining conducted on the prescriptions in the literature to summarize the medication laws of type 2 diabetes mellitus by modern literature based on liver in Traditional medicine.Through the network pharmacology research method,the main active components,targets and action mechanism of the core traditional Chinese medicine from liver for the treatment of type 2 diabetes mellitus were explored using data mining.Methods: The literature on the treatment of type 2 diabetes mellitus based on liver in Traditional medicine was retrieved by using the database.The literature that met the requirements was screened by inclusion and exclusion criteria,and the prescriptions were organized.The data of traditional Chinese medicine involved in the prescriptions were normalized.After the repetitive prescriptions were removed,the information was entered into the TCM inheritance assistance platform and the database was established.The statistics of medication frequency,four-nature-five-taste,and meridian tropism,as well as the formula rule,association rule and clustering analysis were performed through the platform,and the core traditional Chinese medicine and the new prescription were obtained.The active components of core Chinese medicine were obtained through the TCMSP database.The Gene Cards was used to obtain targets for type 2 diabetes,and the common target information of active components of Chinese medicine and disease targets was obtained through the Venny2.1.0 platform.The common target information was input into the DAVID database for GO function annotation and KEGG pathway enrichment analysis.The common target-pathway-disease network relationship diagram was constructed through Cytoscape3.7.1.Results: A total of 197 prescriptions involving 209 Chinese medicines were obtained through literature retrieval.The cumulative use frequency of the drugs was 2,163 times,and the most frequently used drug was Radix Bupleuri,followed by Radix Paeoniae Alba,Poria,Radix Astragali seu Hedysari,and Radix Salviae Miltiorrhizae.The statistical results of nature,taste and meridian tropism were displayed from high to low in frequency of use.The four nature were cold,warm,neutral,cold and hot,and the five taste were sweet,bitter,pungent,sour,salty and acerbity,and the meridian tropism was liver,spleen,stomach,kidney,lung and heart.When the confidence level was 0.9 and the number of support degrees was 40,the formula law was analyzed,The top five drug combinations of the core drug combinations were Radix Paeoniae Alba–Radix Bupleuri,Radix Bupleuri–Poria,Radix Paeoniae Alba–Poria,Radix Paeoniae Alba–Radix Bupleuri–Poria,and Rhizoma Atractylodis Macrocephalae–Radix Bupleuri.The association rules were analyzed and six drug rules were obtained.On the basis of the association rules,the correlation degree was set to 8,and the penalty degree was set to 2 for clustering analysis,and 32 core combinations of new formulae and 16 new formulae were obtained.We conducted network pharmacology research on the core Chinese medicines ranked in the top 5 in terms of use frequency.The main active ingredient of that obtained core chinese medicinal material are linalyl acetate(linalyl acetate),Baicali(baicalin),isorhamnetin(isorhamnetin),kaempferol(kaempferol),quercetin,paeoniflorin(paeoniflorin),hederagenin(hederagenin),Formononetin(formononetin),Mairin(kaempferol),luteolin(luteolin)and others obtained 28 common targets related to diseases,and conducted protein interaction network and network topology analysis on the common targets,obtaining PPARG,AKT1,IL6,VEGFA,TNF and other key targets,and obtaining 108 signaling pathways involving AGE-RAGE signaling pathway,lipid and atherosclerosis,insulin resistance,type II diabetes and other complications of diabetes.Conclusion:(1)The data mining results showed that in the modern literature over the past 20 years,the drug-based taste and nature of the treatment from liver of diabetes mellitus was sweet and cold,and the commonly used core drugs were Radix Bupleuri,Radix Paeoniae Alba,Poria,Radix Astragali seu Hedysari,and Radix Salviae Miltiorrhizae,and the most commonly used drug pair was Radix Paeoniae Alba-Radix Bupleuri.The new prescription drugs have both effects on diabetic with different syndrome types and different symptoms.(2)According to network pharmacology research,it was found that the five core drugs for treatment of diabetes mellitus based on liver participated in multiple pathways,such as insulin resistance,atherosclerosis,diabetic complications and other pathways,to improve glucolipid metabolism and reduce insulin resistance through multiple gene targets such as IL6,NOS3,INSR,STAT3,AKT1,TNF,MTOR,ABCC8,and GCK,to treat T2 DM. |
