Objective:Analyze the effects of Tianniu Zhixuan Capsule on H9C2 myocardial cells and LncRNA H19/miR-675/CaMKⅡ δ signal path.Explore the effect of Tianniu Zhixuan capsule on H9C2 cardiomyocyte hypertrophy model in a certain period of time.Methods:1.Use CCK8 method to determine the adaptive concentration of its Tianniu Zhixuan capsule,and the drug concentration with the best cell survival rate was selected for the experiment.2.Select a certain number of H9C2 myocardial cells and divide them into four groups at random: namely blank serum group,model control group,Tianniu Zhixuan group,and enalapril group.No drugs were added to the blank serum group,and angiotensin II(1.0μg/L)was added to the model control group establish cardiomyocyte hypertrophy model and culture for 24 hours.Detect the m RNA expression of ANP in myocardial cells by real-time quantitative PCR to determine whether the model was successful.Add angiotensin II(1.0μg/L)after the establishment of cardiomyocyte hypertrophy model and culture for 24 hours,add 10% Tianniu Zhixuan capsule medicated serum to culture for 6hours,24 hours and 48 hours respectively.Add angiotensin II(1.0μg/L)to enalapril group after the establishment of cardiomyocyte hypertrophy model to culture for 24 hours,add 10% enalapril medicated serum to culture for 6 hours,24 hours and 48 hours respectively.3.Detect the expression of LncRNA H19 and miR-675 in myocardial cells by real-time quantitative PCR.Detect the expression of CaMKⅡδ in myocardial tissue by Western Blot method.4.Use SPSS and statistical analysis methods to analyze relevant data and draw conclusions.Results:1.The effect of drugs on cell survival: Compared with the medicated serum of Tianniu Zhixuan Capsule of other concentrations,the cell survival rate is the best under the condition of 10% of the medicated serum of Tianniu Zhixuan Capsule.Select this concentration of Tianniu Zhixuan medicated serum for subsequent experiments.2.Gene expression:Compared with the model control group,the expression of LncRNA H19 in myocardial cells of the Tianniu Zhixuan group and enalapril group increased(p<0.05),and the Tianniu Zhixuan group increased significantly(p<0.05).Compared with the model control group,the expression of miR-675 in myocardial cells of Tianniu Zhixuan group and enalapril group increased(p<0.05),and the expression of miR-675 in Tianniu Zhixuan group increased significantly(p<0.05).3.Protein expression:Compared with the model group,the expression of CaMKⅡδ in the Tianniu Zhixuan group and enalapril group decreased(p<0.05),and the decrease in the Tianniu Zhixuan group was more obvious(p<0.05).Conclusion:1.Tianniu Zhixuan Capsule can increase the expression of miR-675 by up-regulating the expression of LncRNA H19 in myocardial cells in vitro,thus down-regulating the expression of CaMKⅡδ.That plays the role in inhibiting the hypertrophy of cardiac myocytes.2.There is no significant difference in the improvement effect of Tianniu Zhixuan capsule on cardiomyocyte hypertrophy model in vitro within a certain period of time.3.Compared with enalapril,the improvement effect of Tianniu Zhixuan capsule on cardiomyocyte cells is more significant on the path of LncRNA H19/miR675/CaMKⅡδ.
