Objective:To verify the efficacy and safety of Tianniu Zhixuan capsule in delaying Left ventricular hypertrophy(LVH).To analyze the effect of Tianniu Zhixuan capsule on the regulation of lncH19 and miR-675 in myocardial tissue of Spontaneously hypertensive rats(SHR)induced by CaMKⅡδ,and to explore the mechanism of Tianniu Zhixuan capsule in reducing blood pressure and delaying left ventricular hypertrophy.Methods: 30 SPF male SHR at 7 weeks of age and 10 WKY rats of the same strain and the same week were selected.After one week of adaptive feeding,10 WKY rats were set as normal control group,and 30 SHRS were divided into model control group,Tianniuzhixuan group and enalapril group according to the basic blood pressure,with 10 rats in each group.The Tianniu Zhixuan group and enalapril group were given corresponding drugs by gavage,respectively.The Tianniu Zhixuan group was given gavage 3 times a day,and the enalapril group was given gavage twice a day for8 weeks.The normal control group and the model control group were given the same amount of distilled water by gavage,3 times a day for 8 weeks.The body weight of rats in each group was measured every Sunday.At the end of the 8th week,the rats were sacrificed to measure the Left ventricular mass(LVM)and calculate the Left ventricular mass index(LVMI).The expression levels of lncH19 and miR-675 in myocardial tissue were detected by Quantitative Real-time PCR(RT-q PCR).The expression level of CaMKⅡδ protein in myocardial tissue was detected by Western blot.Results: 1.Left ventricular mass(LVM)and left ventricular mass index(LVMI): The LVM and LVMI in the Tianniu anti vertigo group and enalapril group were significantly reduced compared to the model control group(P<0.01),and there was no significant difference between the Tianniu anti vertigo group and the enalapril group(P>0.05).2.Protein expression level: Compared with the model control group,CaMK II in the myocardial tissue of the Tianniu anti vertigo group and the enalapril group was observed δ The protein expression levels all decreased(P<0.01).3.Gene expression level: Compared with the model control group,the lncH19 expression in myocardial tissue of the Tianniu Zhixuan group and the enalapril group increased;The expression of miR-675 in myocardial tissue increased in the Tianniu anti dizziness group and the enalapril group.Conclusion: Tianniu Zhixuan capsule can up-regulate the expression of lncH19,up-regulate the expression of miR-675,and down-regulate the expression of CaMKⅡδ protein,which may be related to the reduction of blood pressure and target organ damage.It is one of the important mechanisms of Tianniu Zhixuan capsule in the treatment of hypertensive left ventricular hypertrophy.
