Objective: To study the effect of Xintongtai granules on blood lipids and TXB2,6-KetoPGF1 in AS mice α The impact of expression.Methods: Fifty-two SPF grade male Apo E-/-mice aged 6-8 weeks were selected as the model group,fed with high-fat diet,and 1 2 SPF grade healthy male C57BL/6J wild mice were selected as the blank group,fed with common diet,with an individual weight of20(± 2)g.After 12 weeks of feeding,2 mice were randomly selected from the two groups.After HE staining,the aortic vascular endothelium of mice taken from the model group was observed to have obvious unstable AS plaque,lipid accumulation products,foam cells,etc.The aortic vascular endothelium of mice taken from the blank group fed with ordinary diet was smoother than the former,indicating that the AS model was successful.(Figure 1).After that,the remaining 50 Apo E-/-mice were randomly divided into model group,Xintongtai granule low dose group,middle dose group,high dose group and atorvastatin group with 10 mice each.After being gavaged with normal saline,low,medium and high doses of Xintongtai,and atorvastatin calcium for 8 weeks,the animals were killed,and the experimental samples were selected to detect TC,TG,LDL-C,HDLC by enzymatic method and colorimetric method respectively.The pathological changes of aorta tissue were observed under high power microscope,and plasma TXB2,6-Keto-PGF1 were determined by ELISA α Horizontal.Results: 1.Compared with the model group,each drug group can reduce the serum TC,TG,LDL-C and increase HDL-C of AS mice.The high-dose group of Xintongtai granules is better than the low-dose group(P<0.05).There is no difference between the high-dose group of Xintongtai granules and the statin calcium group(P>0.05).2.Compared with the model group,each drug group can reduce the level of TXB2 in the plasma of AS mice(P<0.05).Xintongtai granule high-dose group and statin calcium group were better than Xintongtai granule low-dose group(P<0.05).3.Compared with the model group,each drug group can make 6-Keto-PGF1 in the plasma of AS mice α The level increased(P<0.05).The high dose group of Xintongtai granules and the atorvastatin calcium group were better than the low dose group of Xintongtai granules(P<0.05).4.Compared with the model group,each drug group can alleviate the pathological changes of aortic vessels in AS mice under the microscope,and the effect of Xintongtai granule high-dose group and atorvastatin calcium group is the most significant.Conclusion: 1.Xintongtai granules can reduce serum TC,TG and LDL-C,and increase HDL-C in AS mice,indicating that Xintongtai granules can intervene the progress of AS by regulating blood lipids.2.Xintongtai Granule can improve the pathological changes of aortic tissue in AS mice,reduce lipid infiltration,and alleviate the formation speed of foam cells and plaques.3.Xintongtai granules can reduce TXB2 levels in the aorta of AS mice and increase 6-Keto-PGF1 levels α Level,reduce TXB2,6-Keto-PGF1 α Ratio,thereby reducing the opportunity for platelet aggregation,thrombosis,plaque formation,and inflammatory response,thereby reducing endothelial injury response and alleviating AS progression.
