Objective: To investigate the effect of Xintongtai Granules on growth factor PDGF-B and VEGF in atherosclerosis mice.Methods: In the experimental group,42 4-week-old male Apo E-/-mice were selected and fed to the SPF laboratory of Hunan University of Traditional Chinese Medicine for high-fat diet.In the blank group,12 wild-type mice were selected and fed with ordinary feed.After 12 weeks of continuous feeding,two mice were randomly selected from the blank group and the experimental group.After HE staining,whether or not the AS model was successfully prepared was determined by whether the aorta of the mouse atherosclerotic plaque appeared.After the model was established,four groups of controls were randomly formed.They were model group,Xintongtai low dose group,Xintongtai high dose group,atorvastatin calcium group,10 in each group.After 8 weeks of oral administration,animals were treated under aseptic conditions.TC and TG were detected by enzymatic method,LDL-C and HDL-C were detected by colorimetric method,serum PDGF-B and VEGF were detected by ELISA,and immunohistochemistry.The PDGF-B,VEGF and Rt-PCR methods were used to detect the m RNA expression of PDGF-B and VEGF in thoracic aorta tissue.Results: 1.Compared with the model group,Xintongtai Granules and statins can significantly reduce TC,TG,LDL-C(p<0.05)and increase HDL-C levels(p<0.05).2.Compared with the model group,the drug group can significantly reduce the serum PDGF-B,VEGF concentrations(p <0.05).3.Compared with the model group,the drug group can down-regulate the positive expression of PDGF-B and VEGF and enhance plaque stability.In the four groups of control experiments,the effect of Xintongtai high dose and statin group was significantly stronger than that of low dose group(p<0.05),However,there was no difference between the high-dose group and the statin group(p>0.05).4.Compared with the blank group,PDGF-B-m RNA and VEGF-m RNA in the aorta of the other groups were significantly different(p<0.05).Conclusion: 1.Xintongtai granules can significantly down-regulate the concentrations of TC,TG and LDL-C in AS mice and up-regulate HDL-C concentrations,indicating that Xintongtai can interfere with the development of AS by regulating blood lipids and stabilizing plaques.2.Xintongtai granule can reduce the content of PDGF-B and VEGF in serum,reduce the inflammatory reaction and delay the development of AS.3.Xintongtai Granule can regulate the positive expression of PDGF-B and VEGF,stabilize the plaque,and achieve the purpose of preventing and treating AS.4.Xintongtai Granule can down-regulate the gene expression of aortic tissue growth factor PDGF-B and VEGF,thus reducing vascular inflammation and achieving anti-AS. |
