Effect Of Jianpi Runfu Decoction On Cellular Inflammatory Factors And Skin Barrier Function In Mice With Atopic Dermatiti

Research purposes:Atopic dermatitis(AD)is a chronic,relapsing,inflammatory skin disease characterized by dry skin,itching,and polymorphic skin lesions.Epidemiological studies over the past 20 years have shown that the prevalence of AD is gradually increasing.In 1998,a survey was conducted on adolescents in 11 provinces and cities in China,and the standardized prevalence rate of AD was 0.69%.In 2002,a survey was conducted on children in 10 cities in China,and the standardized prevalence rate of AD was 3.07%.In 2014,a survey was conducted on children in 12 cities in China,and the standardized prevalence rate of AD was 12.94%.Modern medical research believes that AD is closely related to factors such as skin barrier dysfunction,immune abnormality,skin flora disorder,and family inheritance,but the specific mechanism of action is still unclear.The number of AD patients is gradually increasing.However,there is no clear cure for this disease.Therefore,the effective treatment of AD has become a research hotspot.Skin barrier dysfunction is an important part of the pathogenesis and aggravation of AD,and repairing the skin barrier is one of the important means to prevent the occurrence and development of AD.At present,most AD treatments are emollients,glucocorticoids or biological agents,etc.,long-term use has side effects and infection risks,and is expensive.Therefore,AD treatment methods that are cost-effective,have few side effects,and can repair the skin barrier and reduce skin inflammation have good application prospects.Mr.Zhao Bingnan,a famous doctor of traditional Chinese medicine,believes that this disease is caused by spleen deficiency and blood dryness with dampness,invigorating the spleen and dampness,nourishing the blood and moisturizing the skin,she created the Jianpi Runfu Decoction.The Dermatology Department of Beijing Hospital of Traditional Chinese Medicine conducted a study on the clinical efficacy of Jianpi Runfu Decoction in the treatment of AD,the improvement of skin physiological indicators,and the influence of cytokines.It was confirmed that Jianpi Runfu Decoction can improve the clinical symptoms and skin physiological indicators of patients.This experiment aims to explore the biological evidence that Jianpi Runfu Decoction improves skin barrier function and reduces skin inflammation through animal experiments,and provides a richer theoretical basis for the clinical application of Jianpi Runfu Decoction.Research methods:The scale-tailed rat(Flaky Tail,FT)was used as the AD model mouse.Thirty 20-week-old mice were randomly divided into 6 groups,blank control group,spleen-deficiency model group,high-dose Chinese medicine group,medium-dose Chinese medicine group,low-dose Chinese medicine group,and positive drug loratadine group,with 5 mice in each group.After one week of adaptive feeding,the model of spleen deficiency syndrome was established,and 108mg/ml rhubarb water decoction was administered orally at 0.3ml/d for 1 week;From the third week,Jianpi Runfu Decoction and positive drugs were given by oral gavage Loratadine,Chinese medicine high dose group 1.89g/ml,medium dose group 1.26g/ml,low dose group 0.63g/ml,positive drug loratadine group 0.108mg/ml,0.3ml/d for 2 weeks,and the samples were taken one day after the end of administration.Observation indicators include:the general condition of the mice and the number of times of scratching,the severity score of the skin lesions of the mice,the histopathological manifestations of the mouse skin lesions observed by HE staining,the detection of IL-4,IL-17,IL-22 and TSLP by ELISA.Research result:The positive drug loratadine,high-dose Chinese medicine and middle-dose Chinese medicine can obviously alleviate the skin lesions in mice,while the low-dose Chinese medicine can slow down the progress of skin lesions in mice.HE staining showed that the mice in the blank control group had obvious epidermal hyperplasia,thickened epidermis,and acanthosis;the mice in the high-dose Chinese medicine group,medium-dose group,low-dose group,and positive drug loratadine group had significantly reduced epidermal hyperplasia and cell edema,the thickness of the epidermis was significantly reduced,and with the increase of the concentration of traditional Chinese medicine,the effect was enhanced.The expression levels of IL-4,IL-17,IL-22 and TSLP in skin lesions of high-dose,middle-dose,low-dose and positive drug loratadine groups decreased.Compared with the blank control group,the concentrations of IL-4,IL-17 and TSLP in serum of mice in each Chinese medicine group and positive drug group decreased.In addition to low-dose Chinese medicine group,the concentration of IL-22 in serum of mice in other Chinese medicine group and positive drug group decreased.conclusion:Jianpi Runfu Decoction can effectively improve skin lesions such as erythema,hypertrophy,and dry skin in atopic dermatitis model mice;reduce epidermal hyperplasia and cell edema,and reduce epidermal thickness;down-regulate IL-4,IL-17.IL-22,TSLP levels.Jianpi Runfu Decoction can relieve and treat atopic dermatitis.
