| Purpose:By analyzing the medication rules of professor Wu Wensheng’s prescriptions for treating patients with coronary heart disease,the core drugs were obtained.The network pharmacology method was used to analyze the mechanism of action of the core high-frequency drugs for treating coronary heart disease and explore the correlation between the mechanism of action and iron death mechanism,providing reference value for the prevention and treatment of this disease.Material and method:1.The initial prescriptions of patients diagnosed as "coronary heart disease" or "chest paralysis" by Professor Wu Wensheng in the Cardiology Department of The Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from October 2019 to October 2021 were collected and input into Excel to establish the TCM prescription database,and the frequency analysis of the recorded TCM and its sexual and odor pathway was conducted.SPSS22.0 software and SPSS Modeler 18.0 were used for cluster analysis and network analysis to determine the association combination of high-frequency drugs with drugs.2.On the basis of the above,the main active components and targets of core high-frequency drugs were screened from TCMSP database by network pharmacology method,and target proteins were converted into gene abbreviations by Uniprot;The targets related to coronary heart disease were searched by Genecards and Pharm Gkb databases.Drug-disease target Venn diagram was drawn by R language software.Cytoscape3.8.0 software was used to construct the "compound-target" relationship network diagram.Protein interrelationship network and PPI core network were constructed by using String protein database and Cytoscape3.8.0 software.Finally,R software was used for GO enrichment and KEGG signal pathway analysis of potential targets,and the mechanism of action of core drugs on CHD was analyzed according to the results.Results:1.According to the grading standard,a total of 75 medical records of initial diagnosis met the standard,involving 105 Chinese medicines,with a total frequency of 1092 times,among which 22 medicines were used ≥20 times,and the three drugs with the highest frequency were Chuanxiong,Danshen and Danggui.Among the 105 Chinese medicines involved,according to the four qi classification,the frequency of the use of warm drugs was 410 times,accounting for 37.1%,ranking first.Cold drugs took the second place,using 356 times,accounting for32.22%;The number of ordinary drugs used was 296 times,accounting for 26.79%;The lowest frequency was for thermal drugs(21 times)and cooling drugs(22 times),accounting for 1.99% and 1.90%,respectively.According to the classification of drug taste,the frequency of acid,bitter,sweet,spicy,salty,harsh and light use was 100,510,607,417,55,79 and 62 times,accounting for 5.46%,27.87%,33.17%,22.79%,3.01%,4.32% and 3.39%,respectively,and the frequency of sweet use was the highest.Followed by bitter drugs.According to the statistics,2991 valid data were obtained.Among them,the top 6 were liver,heart,spleen,lung,stomach and kidney,which were 19.86%,18.12%,15.68%,12.57%,10.93% and 9.93%,respectively.2.Cluster analysis was performed for drugs with frequency ≥10 times.According to the results of the tree diagram,when the drugs were divided into 4 clusters,Danshen,Chuanxiong and Danggui were grouped into one cluster,and Keel and Oyster were grouped into one cluster.When it was divided into 9 clusters,Danshen,Chuanxiong,Danggui,Keel and Oyster were still one cluster.Network analysis of drugs with frequency ≥20 times found the strongest correlation between Danshen-Chuanxiong-Danggui.3.Core high-frequency TCM medicines include Tanshinone IIA(tanshinone IIA MOL007154),Luteolin(luteolin MOL000006),beta-sitosterol(β-sitosterol MOL000358),Cryptotanshinone(Cryptotanshinone MOL007088),and 88 core targets for the treatment of coronary heart disease.Among them,CCND1,ESR1,EGFR,FOS,AKT1,PTGS2,JUN,VEGFA,MYC,CASP3,MMP9,STAT3,TP53 are the key targets.4.GO enrichment analysis found that the biological process of core drugs(BP)involves response to metal ion,response to oxygen levels,response to drug and other aspects.Cellular Component(CC)mainly involves the aspects of membrane raft,membrane microdomain,membrane region and so on.Molecular function(MF)mainly focuses on DNA-binding transcription factor binding,RNA polymerase II-Specific DNA-binding transcription factor transcription binding,ubiquitin-like protein ligase binding,ubiquitin protein ligase binding and other aspects.5.164 signal pathways were obtained by KEGG enrichment analysis.Core drugs mainly treat coronary heart disease through PI3K-Akt signaling pathway,Lipid and Atherosclerosis,TNF signaling Pathway,HIF-1 signaling pathway,and IL-17 Signaling pathway,Apoptosis and other related pathways play a role.Conclusion:By analyzing the prescriptions of coronary heart disease patients in Professor Wu Wensheng’s outpatient department,the core high-frequency drugs were found to be Chuanxiong,Danshen and Danggui.It was confirmed that the key active ingredients of the core high-frequency drugs,including tanshinone,luteolin,β-sitosterol and cryptotanshinone,mainly acted on CCND1,ESR1,EGFR,FOS,AKT1,PTGS2,JUN,VEGFA,MYC,CASP3,MMP9,STAT3,TP53 and other key targets.By regulating PI3K-Akt signaling pathway,TNF signaling pathway,lipid and atherosclerosis pathway,IL-17 signaling pathway,HIIF-1signaling pathway,apoptosis and other related pathways,it plays a role in the treatment of coronary heart disease. |
