Purpose:To predict the mechanism of Jiawei Sini San in treating type 2 diabetes mellitus complicated with dyslipidemia by network pharmacology combined with metabonomics technology.Material and method:The mechanism of Jiawei Sini San in treating type 2 diabetes mellitus complicated with dyslipidemia was analyzed by using the technology of network pharmacology,which was divided into the following steps: 1.The effective components and targets of 10 kinds of drugs in Jiawei Sini San,such as Bupleurum,Paeonia alba,immature bitter orange,licorice,Poria,Atractylodes macrocephala,hawthorn,salvia miltiorrhiza,Alisma orientalis and Pueraria lobata,were retrieved through TCMSP database and Uniprot database,and the duplicates were summarized and deleted.2.The disease targets of type 2 diabetes mellitus complicated with dyslipidemia were retrieved through Gene Cards database and OMIM database,and the common targets of drugs and diseases were obtained by intersection of Venn diagram and targets of Jiawei Sinisan.3.Construct the interaction network diagram of "drug-component-target-disease" by Cytoscape3.6.0.4.Using STRING database to construct protein-protein interaction network diagram(PPI),carry out topology analysis,and get the core target of PPI network.5.GO enrichment analysis of key targets and KEGG information pathway and BP biological process network diagram.6.Recruit volunteers with type 2 diabetes mellitus complicated with dyslipidemia,collect blood to make serum samples,use metabonomics technology to get differential metabolites and analyze KEGG enrichment pathway.Results:In this study,the mechanism of Jiawei Sinisan in treating type 2 diabetes mellitus complicated with dyslipidemia was analyzed by network pharmacology.By constructing the network diagram of "drug-component-target-pathway",129 key targets for treatment were obtained.The top ten key targets were JUN,STAT3,RELA,MAPK3,AKT1,MAPK1,IL6,MAPK14,FOS and RXRA.181 action pathways were obtained by GO and KEGG enrichment analysis,and the main pathways were AGE-RAGE signal pathway,TNF signal pathway,PI3K-Akt signal pathway,MAPK signal pathway,HIF-1 signal pathway,etc.Using metabonomics technology,we compared the normal group with the control group,and got 111 differential metabolites.After KEGG enrichment analysis,we got 78 pathways.After crossing the two pathways,we got 18 pathways,among which the key pathways were c AMP pathway and c GMP-PKG signal pathway.Conclusion:The treatment of type 2 diabetes mellitus complicated with dyslipidemia with Jiawei Sini San may be related to its regulation of AGE-RAGE signal pathway and c AMP pathway in oxidative stress,inflammatory reaction and lipolysis.
