Purpose:Explore the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,apply the network pharmacology research method to predict the active components,targets and signaling pathways of high-frequency traditional Chinese medicine in the treatment of diabetic peripheral neuropathy,and build a biomolecular network,from "drug-component" "-target-pathway" multi-level and multi-angle to explore its mechanism of action in the treatment of diabetic peripheral neuropathy,and provide corresponding information and data support and reference for new drug research and development.Material and method:1.Based on the information system of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine,a retrospective analysis of 120 patients who were treated in the Endocrinology Department of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine and the first clinical diagnosis of diabetic peripheral neuropathy were collected.A database was established,and the WPS Office software was used to conduct frequency analysis on drugs,drug types,drug properties,drug flavors,and meridians.Using SPSS Statistics 25.0 software cluster analysis,to explore the medication rules of diabetic peripheral neuropathy.The potential mechanism of high-frequency traditional Chinese medicine was analyzed by means of network pharmacology.2.Use the TCMSP database to retrieve the chemical components of high-frequency drugs,predict protein target information,and establish a high-frequency drug active ingredient-target database.Search Gene Cards,DRUGBANK,OMIM,Disgenet databases for potential targets of diabetic peripheral neuropathy.The drug targets and disease targets were intersected and imported into the STRING database platform to construct a protein interaction(PPI)network.The disease-drug-active ingredient-key target network was constructed using Cytoscape 3.7.2software.Finally,GO enrichment and KEGG pathway enrichment analysis were performed on the targetsResults:1.The medical records of 120 patients were included,covering 4 types of TCM syndromes,2syndrome elements for disease locations and 10 syndrome elements for disease characteristics were extracted.2.The medication rule of diabetic peripheral neuropathy shows that there are 184 traditional Chinese medicines involved,and the commonly used high-frequency traditional Chinese medicines include red peony root,safflower,chuanxiong,angelica,salvia,pinellia,Polygonatum chinensis,dogwood meat,Ophiopogon japonicus,ginseng,chicken Neijin Wait.The most commonly used medicine is tonic,among them,qi 、yin and blood drugs are the main ones,which is mainly warm in nature and flavor,and enters the spleen meridian.Commonly used traditional Chinese medicine(frequency ≥ 20)cluster analysis can be divided into three categories.Clustering prescriptions:(1)Lycium barbarum,Ligustrum lucidum,Anemarrhena,Rhizoma Rhizoma,Hengqi,Ophiopogon japonicus,Polygonatum chinensis,Chinese dogwood meat,Angelica sinensis,ginseng,for the treatment of Xiaozikan drink addition and subtraction(Jin Wenqing recipe);(2): Panax notoginseng,hematoxylin,Radix Glycyrrhizae,Corydalis,Astragalus,Safflower,Chuanxiong,Red Peony,and Salvia are the addition and subtraction of Mudan particles;(3): Achyranthes,Coix Seed,Chicken Neijin,Divine Comedy,Pinellia,Chenpi,Coptidis,Magnolia,Poria,Licorice,are addition and subtraction for Huanglian Wendan Decoction.3.Through TCMSP,Gene Cards,OMIM,Disgenet and other databases,124 active ingredients and 223 targets were screened with the top 5 high-frequency traditional Chinese medicines.Among them,safflower has 72 targets,angelica has 86 targets,salvia has 288 targets,chuanxiong has 36 targets,and red peony has 132 targets.1187 disease targets for diabetic peripheral neuropathy.Obtained 97 high-frequency TCM and disease intersection targets.4.The PPI network mainly involves key target proteins such as AKT1,IL-6,TNF,and VEGFA;GO functional analysis has enriched 132 pathways,which are mainly related to cytokine receptor binding,DNA binding transcription factor binding,receptor ligand activity,and signal receptor binding.Activator activity,RNA polymerase II-specific DNA binding transcription factor binding,cytokine activity,etc.KEGG enriched 168 pathways,including lipids and atherosclerosis,fluid shear stress and atherosclerosis,the role of AGE-RAGE signaling pathway in diabetes complications,PI3K-Akt signaling pathway signaling pathway,and cancer.The proteoglycan may be the main signaling pathway for high-frequency drugs to act.Conclusion:1.The clinical syndromes of diabetic peripheral neuropathy are mainly qi and yin deficiency and blood stasis.In the analysis of syndrome elements,the disease position syndrome elements are mainly meridian,and the disease syndrome elements are mainly blood stasis.The medication rules mostly involve traditional Chinese medicines that are sweet,warm and tonic,nourishing qi and invigorating blood,and most of the meridians enter the spleen and liver meridians.Clustering Fangzhixiaozikanyin confirmed this point of view.2.In this study,the network pharmacology analysis method was used to reveal the active components,gene targets and signaling pathways of high-frequency drugs in the treatment of diabetic peripheral neuropathy.It is preliminarily explained that the therapeutic effect of high-frequency drugs on diabetic peripheral neuropathy may be through anti-oxidative stress,inhibiting the production of pro-inflammatory cytokines,and improving insulin resistance. |
