Study On The Mechanism Of Huangqi Guizhi Wuwu Decoction In The Treatment Of Diabetic Peripheral Neuropathy Dased On Network Pharmacology

Purpose: Through network pharmacology technology,explore the target points and signal pathways of Huangqi Guizhi Wuwu Decoction in the treatment of diabetic peripheral neuropathy,and explore its possible mechanism of action.Methods: Based on the Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP),search for the chemical components and in vivo targets of Huangqi Guizhi Wuwu Decoction,and mine through the human gene database(Gene Cards)and the online human Mendelian genetic database(OMIM)Potential target of diabetic peripheral neuropathy(Diabetic Peripheral Neuropathy,DPN).Construct Protein-Protein Interaction Networks(PPI)network through STRING database.Cytoscape 3.7.2 software was used to construct a drug-compound-target-disease network,and the Bioconductor bioinformatics software package was used for GO function enrichment and KEGG pathway analysis.Results: A total of 8 active ingredients related to DPN,6 directly related targets,and 9related metabolic pathways were discovered through the above methods.The key ingredients are quercetin,luteolin,kaempferol,baicalein,β-carotene,ginsenoside rh2,β-sitosterol,strychnine,etc.;the targets related to DPN are PTGS2,CASP3,JUN,MMP9,AKT1,TNF;related metabolic pathways are Fluid shear stress and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,Chagas disease(American trypanosomiasis),TNF signaling pathway,Hepatitis B,Kaposi sarcoma-associated herpesvirus infection,Human cytomegalovirus infection,Proteoglycans in cancer,Human immunodeficiency virus 1 infection.Conclusion: Huangqi Guizhi Wuwu Decoction exerts its anti-oxidation,anti-inflammatory,anti-apoptosis,nerve regeneration and neuroprotective effects through a multi-component,multi-target,and multi-pathway synergistic manner.It may be the underlying mechanism of Huangqi Guizhi Wuwu Decoction for the treatment of DPN.
