Purpose:To observe the effect of Jianpi Huayu Decoction on NF-κB related protein in chronic atrophic gastritis model rats,and to explore the therapeutic effect and mechanism of Jianpi Huayu Decoction on chronic atrophic gastritis.Material and method:In this experiment,a completely randomized design was adopted.60 SPF SD rats were randomly divided into 15 in the normal group(ZC group)and 45 in the model group.After 3 days of adaptive feeding,the rats in the modeling group were given 150μg/ m L N-methyl-N-nitrosoguanidine(N-methyl-N’-Nitro-n-nitrosoguanidine,MNNG) solution for free drinking and 0.03% ranitidine diet for free consumption from the 4th day.After 2 days of satiety and 1 day of fasting,the rats in the modeling group were given 40% ethanol 1m L /100 g for intragastric feeding for 12 weeks.During the whole experiment,the normal group was fed with normal feed water.After pathological verification,the remaining CAG rats were divided into model blank group(MX group)(n =11),Chinese medicine treatment group(ZY group)(n =12),Western medicine treatment group(XY group)(n =11)according to the random number table.The ZY group was given Jianpi Huayu Decoction(9.27 g/kg crude drug),the XY group was given Vitacoenzyme(0.27 g/kg),and the MX group was given equal dose of double steam water.After 12 weeks of treatment,all rats were sacrificed and sampled.Observation indicators: The general status of rats was recorded during the experiment,gastric mucosa of rats and gastric pathological changes of rats in each group were observed after the experiment.The expression levels of key proteins p65 and p-p65 and downstream factor COX-2 in NF-κB signaling pathway of gastric mucosa were detected by Weston blot method.SPSS 26.0 was used to analyze the data.Results:1.General state: Compared with ZC group,MX group rats showed decreased water intake,slower weight gain,abnormal behavior and dark yellow fur.After treatment with Jianpi Huayu Decoction,the general state of ZY group was significantly improved compared with MX group.2.Gastric mucosa of rats: Compared with ZC group,the overall color of gastric mucosa of MX group was dim,with red and white,and some blood vessels were visible,scattered in hard grayish white nodules.The gastric mucosa status of ZY group was improved compared with MX group after Jianpi Huayu Decoction treatment.3.Gastric mucosa pathology: Compared with ZC group,gastric mucosa atrophy was more severe in MX group(P<0.05),and the atrophy score was higher(P<0.05),showing statistical differences.Compared with MX group,the degree of atrophy in ZY group and XY group was significantly decreased(P<0.05),and the atrophy score was decreased(P<0.05),showing statistical difference.There were no significant differences in the proportion and score of atrophy between ZY group and XY group(P>0.05).4.The protein expression levels of NF-κB p65,NF-κB p-p65 and COX-2 in gastric mucosa tissue: Compared with ZC group,the protein expression levels of NF-κB p65,NF-κB p-p65 and COX-2 in MX group were increased(P<0.05),with statistical difference.Compared with MX group,the protein expression levels of NF-κB p65,NF-κB p-p65 and COX-2 in ZY group and XY group were decreased(P<0.05),showing statistical differences.There were no significant differences in NF-κB p65,NF-κB p-p65 and COX-2 protein expression between ZY group and XY group(P>0.05).Conclusion:1.Jianpi Huayu Decoction can treat CAG,improve gastric mucosa atrophy and inflammation.2.The mechanism of Jianpi Huayu Decoction in improving gastric mucosa atrophy and inflammation of CAG may be related to inhibiting NF-κB and decreasing COX-2 expression. |
