Effects Of “Kaixuan Xiezhuo Recipe” On The Balance Of Th17/Treg And Related Cytokine Expressions In Peripheral Blood Of Rats With Yang Deficiency Psoriasi

Objective: To investigate the effect of Kaixuan Xiezhuo Decoction on Th17/Treg balance and expression of related cytokines in peripheral blood of Yang deficiency psoriasis rats.Methods: 70 SD rats were randomly divided into blank group and model group.After depilation,15 rats in the blank group were smeared with Vaseline on the back skin of rats,and 55 rats in the model group were gavaged with 100% Rhubarb decoction for 14 days.At the same time,5% imiquimod cream was smeared on the back skin of rats once a day for 2 weeks,and the 14 th day of modeling.After successful modeling,the rats were divided into blank control group,model control group,Tripterygium glycoside tablet group and Kaixuan Xiezhuo Decoction low,medium and high dose groups,with10 rats in each group.The blank control group and model control group were gavaged with normal saline once a day,Tripterygium glycoside tablet group was gavaged with Tripterygium glycoside tablet solution once a day,and Kaixuan Xiezhuo Decoction low,medium and high dose groups were gavaged with Kaixuan Xiezhuo decoction solution of different concentrations once a day for 7 days.24 hours after the last administration,the skin lesions on the back of rats were scored by PASI.Then the abdominal aorta of rats in each group was collected with 8ml blood,the ratio of Th17/Treg was detected by flow cytometry,and IL-17 and TGF-β were detected by ELISA.The skin of the back was taken for pathological section.Results:(1)The PASI scores of model control group,Tripterygium glycoside tablet group and Kaixuan Xiezhuo Decoction low,medium and high dose group were significantly different from those of blank control group(P<0.05),and the PASI scores of Tripterygium glycoside tablet group,Kaixuan Xiezhuo Decoction low,medium and high dose group were significantly different from those of model control group(P<0.05);(2)The histopathological changes of back skin lesions in Tripterygium glycoside tablet group and Kaixuan Xiezhuo Decoction low,medium and high dose group were lighter than those in model control group;(3)The ratio of Th17/Treg in the model control group was 1.31 ±0.38 and that in the blank control group was 0.30 ±0.17.The ratio of Th17/Treg in the model control group was significantly higher than that in the blank control group(P<0.05);The Th17/Treg ratios of Tripterygium glycoside tablet group and Kaixuan Xiezhuo Decoction low,medium and high dose groups were 0.67 ±0.12,0.75 ±0.23,0.67 ±0.12 and 0.45 ±0.10 respectively,which were significantly lower than those of the model control group(P<0.05);(4)The level of IL-17 in the model control group was 29.40 ±6.00ng/ml and that in the blank control group was3.31±1.33ng/ml.The level of IL-17 in the model control group was significantly higher than that in the blank control group(P<0.05);The levels of IL-17 in Tripterygium wilfordii polyglycoside tablet group and Kaixuan Xiezhuo Decoction high,medium and low dose groups were 11.19 ±1.23ng/ml,17.22 ±2.16ng/ml,7.58 ±1.50ng/ml and5.52±0.23ng/ml respectively,which were significantly lower than those in the model control group(P<0.05).The level of TGF-β in Model control group was 9.09 ±1.57ng/ml in the model control group and 4.39 ±1.75ng/ml in the blank control group.The level in the model control group was higher than that in the blank control group(P<0.05);The levels of TGF-β in Tripterygium glycoside tablet group and Kaixuan Xiezhuo Decoction low,medium and high dose groups were 14.50 ±1.54ng/ml,11.35 ±0.48ng/ml,18.12 ±1.75ng/ml and45.49 ±3.47ng/ml respectively,which were higher than those in the model control group(P< 0.05).Conclusion:(1)It can be observed that the PASI score of psoriasis like lesions in SD male rats in the low,medium and high dose groups of Kaixuan Xiezhuo Decoction is lower than that in the model control group,suggesting that Kaixuan Xiezhuo Decoction has a certain effect on psoriasis like lesions;(2)The ratio of Th17/Treg in the model control group was higher than that in the blank control group,suggesting that the pathogenesis of psoriasis may be related to the imbalance of Th17/Treg ratio;(3)The mechanism of Kaixuan Xiezhuo Decoction in the treatment of Yang deficiency psoriasis may be to reduce the level of Th17 cells and increase the level of Treg cells by regulating the ratio of Th17/Treg,so as to reduce the expression of related cytokines IL-17 and increase the expression of TGF-β,and the curative effect of high-dose group was the most significant.
