BackgroundAs one of the common digestive diseases in humans,it is a chronic non-specific inflammatory bowel disease caused by many factors and whose cause is still unclear.The factors that cause ulcerative colitis include:intestinal flora imbalance,adaptability Immune disorders,environment and genetics.Ulcerative colitis repeatedly affects the health and quality of life of patients,and its ulcerative colitis-associated colorectal cancer accounts for 9%-11%of deaths in UC patients due to its recurrent episodes.Therefore,ulcerative colitis is The World Health Organization is listed as one of the modern refractory diseases.The number,composition and distribution of human intestinal flora are inextricably linked to intestinal diseases and related metabolic diseases.The intestinal flora of patients with ulcerative colitis can be disordered,and the number and structure of the intestinal flora can also reverse the intestinal tract,further affecting the function of the normal function in the intestinal tract.In recent years,more and more studies have used molecular biology methods to explore the changes in the composition and structure of the intestinal flora in patients with ulcerative colitis,and further on the characteristics of TCM syndromes and intestinal flora in ulcerative colitis.There are relatively few studies.In this paper,16S rDNA high-throughput sequencing technology and bioinformatics analysis method were used to analyze the difference of cold and heat in the active stage of ulcerative colitis,and the difference and characteristics of intestinal flora between normal people.Objectives:To compare and analyze the differences in the composition and structure of intestinal flora between patients with active ulcerative colitis,cold and heat syndrome,and healthy people,and to explore the composition and structure of intestinal flora in patients with active ulcerative colitis and healthy people.The relationship between changes,as well as the composition of intestinal flora and structural changes between different cold and heat in patients with cold and heat syndrome.Method:Experiment one:This study collected 14 patients with spleen,stomach,liver and gallbladder outpatient and inpatient active ulcerative colitis from October 2017 to February 2018,and 14 healthy controls.Collected stool samples and extracted intestinal bacteria.The total DNA of the intestinal flora of the individual specimens was sequenced by high-throughput sequencing technology,and the 16S rDNA V3-V4 variable region was sequenced and analyzed for biological information(classification,abundance analysis and diversity analysis of intestinal flora species).The difference in flora between patients with active ulcerative colitis and normal people was obtained.Experiment two:This study collected 39 patients with spleen,stomach,liver and gallbladder outpatient and inpatient active ulcerative colitis from March 2018 to December 2018,including 19 patients with partial cold syndrome and 20 patients with partial heat syndrome.The patient’s stool specimens were used to extract the total DNA of the intestinal flora of the intestinal flora,and the 16S rDNA V3-V4 variable region was sequenced by high-throughput sequencing technology,and the biological information was analyzed(intestinal flora classification,abundance)Degree analysis and diversity analysis),the difference in flora between cold and heat groups was obtained.Results:Experiment one:(1)There were significant differences in abundance,diversity and structure between the active ulcerative colitis patients and healthy people(P<0.05).(2)Bacteroidetes and Firmicutes in patients with ulcerative colitis at the level of the portal are lower than those in healthy people,but Proteobacteria and sputum(Verrucomicrobia)is higher than healthy people;(3)In the healthy population of the genus,Bacteroides,Roseburia,lachnospira,Prevotella 9 and the like are dominant;patients with ulcerative colitis are mainly Akkermansia,Bifidobacterium,Lactobacillus,Escherichia-Shigella and the like.Experiment Two:(1)There was no significant difference in the abundance and diversity between the two groups in the cold-heat syndrome of the patients with active colitis in the active stage of ulcerative colitis(P>0.05),but compared with the proportion of some strains.Significant difference(P<0.05);(2)The difference in the level of the door may be caused by the proportion of the proteobacteria,and the partial heat may be caused by the proportion of the micro-bacteria;the difference between the genus and the cold group may be Escherichia-Shigella..The proportion of the Hist-Shigella species is dominant,and the partial heat group may be dominated by Bacteroidales.(3)Bacteroidetes,Bacteroidia,Bacteroidales,Eisenbergiella,Hungatella,mainly in the intestines of patients with cold and heat syndrome.In the partial cold group,Proteobacteria,Gammaproteobacteria,Enterobacteriales,Enterobacteriaceae,Escherichia-Shigella,Actinobacteria,Bifidobacteriales,Bifidobacteriaceae,Bifidobacterium,Verrucomicrobia,Verrucomicrobiae,Verrucomicrobiales,Verrucomicrobiaceae,Akkermansia,etc.Conclusion:(1)There was a difference in the abundance,diversity and structure of intestinal flora between active ulcerative colitis patients and healthy controls(P<0.05);in the intestinal tract of patients with active ulcerative colitis It is mainly represented by Actinobacteria represented by Bifidobacterium,Verrucomicrobia represented by Akkermansia,and Escherichia-Shigella.In the healthy control group,Bacteroidetes and other related bacteria such as Firmicutes and Prevotella represented by lachnospira are mainly expressed;(2)There was no significant difference in the abundance and diversity between the two groups in the active stage of ulcerative colitis and cold-heat syndrome.(P>0.05),but the proportion of some strains was compared.Significant difference(P<0.05):There were differences in intestinal flora level and genus level.The difference in door level may be dominated by Proteobacteria,and the partial heat group may be dominated by Bacteroidetes;The difference in genus level may be dominated by Escherichia-Shigella,and the partial heat group may be dominated by Bacteroidales.(3)In the active period,the cold and hot syndrome of the cold and hot syndrome of ulcerative colitis may have different species representing the cold and heat.The representative genus of the cold group is Escherichia-Shigella belonging to the Proteobacteria.The representative genus of the group is Bacteroidales of Bacteroides.
