Objective(s):The structure and diversity of intestinal flora of UC patients and their healthy compatriots were analyzed to provide a scientific basis for digging new therapeutic targets and lay a theoretical foundation for microbial diagnostic targets of UC.Methods:The feces of 50 patients with UC and their healthy compatriots were collected,and 16S rRNA sequencing was used to analyze the biological information of the sequencing results.Results:1.Species note:At the phylum level,the flora is mainly composed of Actinobacteria,Bacteroidetes,Firmicutes and Proteobacteria,At the genus level,the flora is mainly composed of Bacteroides,Lactococcus,Faecalibacterium,Psychrobacter.2.The operational taxonomic units(OTUs)of UC and the control group were 1126,among which 887 were in the UC group and the healthy sib group,485 were exclusive to UC patients,and 641 were exclusive to the healthy sib group.3.Diversity comparison:The a diversity,β diversity and flora abundance of UC group were significantly lower than their healthy sibs.4.Comparison of UC and control group flora:At the phylum level,Proteobacteria and Actinobacteria in UC were significantly increased,while Firmicutes and Bacteroidetes were significantly decreased.At the level of the family,Among UC,Moraxellaceae,Enterobacteriaceae,Ruminococcaceae,Streptococcaceae,Enterococcaceae,and BifidobacteriumBifidobacteriaceae was significantly higher than the control group,while Bacteroidaceae and Peptostreptococcaceae were significantly lower than the control group.At the genus level,In UC,Enterococcus,Streptococcus,Faecalibacterium,Escherichia-Shigella,and Psychrobacter are all species),while Bacteroides,Parabacteroides,Romboutsia and Erysipelatoclostridium were significantly decreased.At the species level,the flora with significantly increased UC were:Streptococcus_parauberis,Streptococcus gallolyticus-subsp-macedonicus,Streptococcus mutans,Streptococcus_parauberis,distant chainStreptococcus_sobrinus,Streptococcus_varani,Escherichia-coli,and Psychrobacter-pulmonis,the flora with significantly reduced UC were as follows:Bacteroides-Caecimuris,Bacteroides-Dorei,Bacteroides-Fragilis,Bacteroides-Salyersiae,manyBacteroides-theTaiotaomicron,Bacteroides_Ovatus,Parabacteroides-Goldsteinii.5.Linear Discriminant Analysis Effect Size(LEFSE)indicated that there were significant differences in the flora of UC group:Streptococcus family,Enterococcus family,Enterobacteriaceae;The species with significant difference in the flora of the control group were tannins and gastrostreptococcus.6.The following bacteria are the fecal flora of UC patients:Salyersiae,Parabacteroides-Goldsteinii,Romboutsia,Psychrobacter-pulmonis,were not reported in a previous study the relationship between the bacterial flora and UC,including salyersiae,Parabacteroides-goldsteinii,Romboutsia has implications to the intestinal inflammation,decreased significantly in UC,and Psychrobacter-pulmonis increases,more than bacterial flora changes may indicate inflammation in patients with UC,for UC microbial target provides the theory basis for the diagnosis and treatment.Conclusion(s):1.In this study,the influence of confounding factors such as genetics and diet was excluded.The results indicated that intestinal flora of UC and the control group were significantly different,and the diversity and abundance of fecal flora of healthy siblings were higher than those of UC group.2.Compared with healthy siblings,Bacteroidetes and Firmicutes were significantly decreased in the UC group,and the above bacteria mainly produced short-chain fatty acids.Linear Discriminant Analysis Effect Size(LEFSE)was used to analyze the Effect Size.The results indicated that the change of intestinal flora in UC patients was mainly the increase of harmful bacteria,accompanied by the decrease of beneficial bacteria.
