bran,bagasse and the other agricultural products).The potency substance in the fermentation process not only provides nutrients such as carbonitride which fungal requires,the enzyme which is produced by the fungus can also change the component of herbs,achieve better efficacy,produce new nature and flavor,function,and reduce the toxicity of raw herbs.There are lots of studies about Ganoderma lucidum.It is a commonly used medicinal fungus.The liquid deep fermentation of Ganoderma lucidum has a short cycle,good product quality,high production efficiency,easy to control and other characteristics,widely used in industrial production applications.Gynostemma pentaphyllum Makin,tartary buckwheat and Cassia obtusifolia L.is medicinals and edible traditional Chinese medicines.They have good hypolipidemic effect,and their corresponding hypolipidemic constituents Gynostemma Pentaphyllum Makin saponins,buckwheat flavonoids and Cassia obtusifolia L.anthraquinones are studied relatively mature.In this paper,Ganoderma lucidum and traditional Chinese medicine Gynostemma pentaphyllum Makin.,Cassia obtusifolia L.and buckwheat were used as research carriers.Optimize the deep fermentation process of Ganoderma lucidum and determine the end of fermentation.Establish bidrectional-fermentation system of traditional Chinese medicines and Ganoderma lucidum.And use pharmacodynamic model to explore whether the traditional Chinese medicine and Ganoderma lucidum fermentation products have good pharmacological effects.In order to better reveal the interaction between medicinal fungus and traditional Chinese medicine and the basic changes in the law.Provide new research ideas for the bidrectional-fermentation technology.The methods and results of this study are as follows:(1)Establish the deep fermentation system of Ganoderma lucidum.Ganoderma lucidum biomass as the standard,to deep fermentation of Ganoderma lucidum.Determine the end of fermentation for 7d.Orthogonal design experiment was used to optimize the extraction process of Ganoderma lucidum polysaccharide and Ganoderma lucidum triterpene.And the method of analysis of polysaccharide and triterpenoids was established.The carbon source and nitrogen source of Ganoderma lucidum deep fermentation medium were screened by single factor test.And the response surface methodology(RSA)was used to control the root of Ganoderma lucidum biomass,polysaccharide and triterpene.According to Box-Behnken center combination design principle,the contents of carbon,nitrogen,KH2PO4 and MgSO4 in the deep fermentation medium of Ganoderma lucidum were optimized.The optimized medium was sucrose 26.57g,mixed nitrogen source 35.00g,MgSO4 1.00 g,KH2PO4 0.50 g.The results of the validation medium were reliable.(2)The method of single factor experiment was used to optimize the extraction conditions of active ingredients of traditional Chinese medicine.The bidrectional-fermentation system of traditional Chinese medicine and Ganoderma lucidum was established.HPLC and colorimetric methods were used to determine the intracellular polysaccharides,intracellular triterpenes,extracellular polysaccharides,extracellular triterpenes;Gynostemma pentaphyllum Makin.Saponin,tartary buckwheat flavonoids and Cassia obtusifolia L.anthraquinone were used to determine the effect of different traditional Chinese medicine on the deep fermentation system of traditional Chinese medicine and Ganoderma lucidum.The results showed that the growth of Ganoderma lucidum could increase the growth of Ganoderma lucidum and increase the production of Ganoderma lucidum and Ganoderma lucidum triterpene,and the high concentration could inhibit the growth and metabolism of Ganoderma lucidum.Tartary buckwheat in the added amount of 1 to 20g/L has a good role in promoting the growth and metabolism of Ganoderma lucidum in the buckwheat addition of more than 20g/L when the inhibition of Ganoderma lucidum growth and metabolism.At the same time,the bidrectional-fermentation of traditional Chinese medicine active ingredient gypenosides cannot be detected because the amount of oligosaccharides cannot be detected,but the Chinese medicine Gynostemma and buckwheat in the rutin and Cassia obtusifolia L.free anthraquinone components in the role of Ganoderma lucidum conversion.(3)To establish a method for the determination of lipase,and to determine the optimum reaction conditions for lipo1 under conditions of 37℃ and pH=8.0 for 15 min.Through the lipase inhibition test,test whether the bidrectional-fermentation system has hypolipidemic effect.The inhibitory effects of different concentrations of fermentation broth on lipase were determined by the optimum reaction conditions of lipase.The results showed that the fermentation broth of three Chinese medicines and Ganoderma lucidum had a good effect on the inhibition of lipase.The results showed that the change of lipase inhibition rate was mainly caused by the concentration of Ganoderma lucidum Exogenous polysaccharides,triterpenes and active ingredients of traditional Chinese medicine rutin or free anthraquinone content changes caused by the higher Chinese medicine in the amount of crude drug,lipase inhibition rate is mainly by the fermentation of traditional Chinese medicine active ingredients. |
