Dalbergiae Odoriferae Lignum(Jiangxiang)is the dry heartwood of the trunk and root of Dalbergia odorifera T.Chen.It can remove blood stasis and stop bleeding,regulate qi and relieve pain,and it can be used to treat hematemesis,traumatic bleeding,liver depression,hypochondriac pain,vomiting and abdominal pain.Modern studies have shown that the main chemical components of Jiangxiang are volatile oil,flavonoids and benzofurans,which have anti platelet aggregation,anti thrombosis,anti-oxidation,anti-inflammatory,anti-tumor and other pharmacological effects.Dalbergia odorifera(rosewood)is also a valuable material for mahogany furniture and handicrafts.Due to the sharp decrease of resources,the sources of Jiangxiang in the market are complex and there are many counterfeits.The current Pharmacopoeia standards lack of specific identification and quantitative control index components,which can not effectively monitor the medicinal materials and preparations.In this paper,the chemical constituents of Jiangxiang were further studied to provide a scientific basis for the formulation of specific quality standards.By using silica gel column,ODS,MCI,gel Sephadex LH-20 and preparation liquid equal separation and purification technology,IR,1D,2D-NMR,ESI-MS and other spectroscopic methods,54 compounds,including 9 new compounds,were isolated and identified from petroleum ether and dichloromethane of Jiangxiang.All the compounds were composed of 18 sesquiterpenoids: dalodorin C(1),dalodorin D(2),dalodorin E(3),6-methyl-2-(5-methyl-5-vinyltetrahydrofuran-2-yl)hept-5-en-2-ol(4),rel-(3S,6R,7S)-3,7,11-trimethyl-3,6-epoxy-1-dodecen-7,11-diol(5),dalbergene(6),dalodorin F(7),(E)-β-farnesol(8),dalodorin G(9),rel-(3R,6R,7S)-3,7,11-trimethyl-3,7-epoxy-1,10-dodecadien-6-ol(10),α-hydroxyclonolidol(11),β-eudesmol(12),pteranol(13),eudesm-4(14)-ene-3α,11 diol(14),α-costol(15),nerolidol(16),2,6,10-trimethyl-3,6,11-dodecatriene-2,10-diol(17),cabreuva oxide(18),among which compounds 1,2,3,7 and 9 are new compounds;There were 24flavonoids: 1,2,4-Trimethoxy-5-(1-phenyl-2-propen-1-yl)benzene(19),R(+)-dalbergiphenol(20),Latifolin(21),R(-)-5-O-methyllatifolin(22),3’-hydroxy-2,4,5-trimethoxydal-bergiquinol(23),3,4-dimethoxydalbergione(24),dalbergin(25),variabilin(26),medicarpin(27),(3R)-vestitone(28),lespedeol C(29),(3R)-4’-Methoxy-2’,3,7-trihydroxyisoflavanone(30),sativanone(31),5,7-dihydroxy-2’,3’,4’-trimethoxyisoflavanone(32),2’,7’-Dihydroxy-4’-methoxyisoflavone(33),formononetin(34),biochanin A(35),2’-methoxybiochanin A(36),5,7,4’-trihydroxy-2’-methoxyisoflavone(37),liquiritigenin(38),isoliquiritigenin(39),7-(2,4-dimethoxyphenyl)-8-(2’,4’-dihydroxyphenyl)-9-propen-8-one(40),mucronulatol(41),7-hydroxy-2’,4’-dimethoxyisoflavan(42),Among them,compound 40 is a new compound;There are 6 benzofurans: 2,3-dihydro-6-methoxy-3-methyl-2-phenyl-5-benzofuranol(43),parvifuran(44),pterolinus D(45),phenylbenzofuran A(46),phenylbenzofuran B(47),2’,6-dihydroxy-4’-methoxydihydroauronol(48),Compounds 45,46 and 47 are new compounds;There are six other classes:3-O-acetyloleanolic acid(49),β-sitosterol(50),stigmasterol(51),melanin(52),pterostilbene(53),4-methoxy-6-hydroxybenzaldehyde(54).Among them,26compounds(6,8,11-18,24,27,30,31,36-39,42-44,48,51-54)were isolated from this plant for the first time.In addition,the anti-inflammatory activities of new compounds and known phenols with similar structure types were studied.The results showed that compounds 40 and 45 could inhibit the release of NO from raw 264.7cells.Compounds 46-47 also had a slight inhibition on the release of NO from raw264.7 cells,indicating that these flavonoids and benzofurans had certain anti-inflammatory activity,while sesquiterpene compounds 1-3,7 and 9 had no anti-inflammatory activity.
