The heartwood of Dalbergia odorifera T.Chen(Jiangxiang)has been widely used to treat cardiovascular and cerebrovascular diseases in China.It can not only promote blood circulation for removing blood stasis but also regulating qi-flowing for relieving pain.It is mainly composed of Zhenggu liniments,Guanxin danshen tablets,Danxiang Guanxin injections and other preparations.Results of modern studies showed that the main components of Jiangxiang are volatile oil and flavonoids,which are proved a variety of biological activities,such as anti-platelet aggregation,anti-thrombotic,anti-oxidation,anti-inflammatory,anti-tumor and other effects.Recently,the resources reduce rapidly after Dalbergia odorifera T.Chen becomes the most expensive mahogany.The sources of it are complex and there are many counterfeits,which brings hidden trouble to the safety of drug use.In the 2015 edition of the Chinese Pharmacopoeia(Volume 1),the standard of Jiangxiang lacks the specific qualitative and quantitative control components.so it will lose the effective monitoring of medicinal materials and their preparations.Therefore,in order to control the quality of Jiangxiang,this study aimed to research their chemical compounds.The results will provide scientific basis for improving the quality standard of Jiangxiang and its preparations.The constituents were extracted,isolated and purified from 95% ethanolic extract of Dalbergia odorifera T.Chen by using various chromatographic techniques such as silica gel column,ODS,MCI,Sephadex LH-20 and High Performance Liquid Chromatography.The structures were identified by spectroscopic analysis including IR,1D and 2D NMR techniques and HR-ESI-MS.We acquired 34 components including 11 sesquiterpenoids,12 flavonoids,3 pterocarpans,2 benzofurans,2 triterpenes and 5 other compounds.The structures of 11 sesquiterpenoids were identified as(3R,9R)-7-hydroxym ethyl-3,11-trimethyl-1,6,10-triene-3,9-diol(1),(3R,6S,7S)-3,7-dimethyl-10-hydroxym ethyl-10-keto-3,7-epoxy-1-alkenyl-6-ol(2),rel-(3S,6R,7R)-3,7,11-tetramethyl-1,9-die ne-3,7-epoxy-6,11-diol(3),(E)-nerolidol(4),6α-hydrocyclonerolidiol(5),rel-(3S,6R,7S)-3,7,11-trimethyl-3,6-epoxy-1,10-dodecadien-7-ol(6),(2Z,6E)-farnesol(7),(3R,6R,7S)-3,7,11-trimethyl-3,7-epoxy-1,10-dodecadien-6-ol(8),rel-(3S,6R,7S)-3,7-dim ethyl-3,6;7,10-diepoxy-1-decanen-10-one(9),(3S,6R,7S,10S)-2,6,10-trimethyl-3,6;7,10-diepoxy-11-dodecen-2-ol(10),rel-(3S,6R,7S,10R)-2,6,10-trimethyl-3,6;7,10-diepox y-11-dodecen-2-ol(11).Among which constituents 1,2,3 are new compounds.The structures of 15 flavonoids were identified as pinocembrin(12),biochanin A(13),galangin(14),sativan(15),angolensin(16),homoferreirin(17),5,7-dihydroxy-2?,3?,4?-trimethoxyisoflavanone(18),7-hydroxy-5?-methoxyspiro[benzo[d][1,3]-dioxole-2,3-chroman]-4-one(19),R(+)-dalbergiphenol(20),3?-hydroxy-2,4,5-trime-t hoxy-dalbergiquinol(21),R-(-)-latifolin(22),5-Omethyllatifolin(23),medicarpin(24),coumestrol dimethyl ether(25),(6a S,11 a S)-homopterocarpin(26).2 benzofurans:(2R,3R)-obtusafuran(27),5-hyroxy-6-methoxy-2-methyl-3-phen ylbenzofuran(28);2 triterpenes: lupeol(29),3β-acetoxyolean-12-en-28-oic acid(30)and 4 other compounds: 3,4-dimethoxydalbergione(31),pterostilbene(32),2,4-dihydroxy-5-methoxy-Benzophenone(33),2-hydroxy-3-methoxyxanthone(34).The sesquiterpenoids 1~6,8~11 were screened for anti-tumor activities on Hela cells,A549 cells and MGC-803 cells by CCK8 method.The results showed that all co mpounds had no significant inhibitory effect on this three tumor cells.In addition,by TLC preliminary identification,developing agent and durable investigation,it was found that the sesquiterpenoids(E)-nerolidol and rel-(3S,6R,7S)-3,7,11-trimethyl-3,6-epoxy-1,10-dodecadien-7-ol and the flavonoids pinocembrin and medicarpin can be used for TLC identification of Dalbergia odorifera T.Chen.
