| Objective: To study the expression and clinical significance of HMGB1 and NF-κB in gastric cancer and adjacent tissues,and to explore the effects of HMGB1 and NF-κB on the biological behavior of gastric cancer cells.Methods: Firstly,the expression of HMGB1 and NF-κB in gastric cancer was analyzed by bioinformatics.Gastric cancer tissues and related paracancer tissues of primary gastric cancer patients underwent surgical resection in the Department of Cancer of Gansu Provincial People’s Hospital from January 2018 to December 2018 were collected.A total of 60 gastric cancer patients meeting the requirements were screened,and the expressions of HMGB1 and NF-κB were detected by immunohistochemistry.The correlation of HMGB1 and NF-κB with clinicopathologic data of gastric cancer patients was analyzed.The effects of HMGB1 and NF-κB expression on the survival of gastric cancer patients were analyzed by the survival curve.The expression of HMGB1 and NF-κB in gastric cancer cells and normal gastric epithelial cells was detected by RT-q PCR.After the expression of HMGB1 was inhibited by glycyrrhizic acid,the expression of NF-κB was detected by RT-QPCR,and the biological behavior of gastric cancer cells was detected by CCK8,wound healing and Transwell.Results: The results of bioinformatics analysis indicated that HMGB1 and NF-κB were highly expressed in gastric cancer,and the expressions of HMGB1 and NF-κB in gastric cancer tissues were significantly higher than those in paracancer tissues.By analyzing the correlation between the expression of HMGB1 and NF-κB and clinicopathologic data,it was found that the expression of HMGB1 and NF-κB was correlated with tumor size,tumor invasion depth,lymph node metastasis and TNM stage,but not with gender,age and distant metastasis of patients.In addition,the study analyzed the relationship between the survival rate of gastric cancer patients and the expression of HMGB1 and NF-κB,and found that the survival time of gastric cancer patients in the low expression group of HMGB1 and NF-κB was significantly longer than that in the high expression group.In terms of biobehavioral effects of HMGB1 and NF-κB on gastric cancer cells,both HMGB1 and NF-κB can promote proliferation,migration and invasion of gastric cancer cells.Conclusions: 1.HMGB1 and NF-κB are highly expressed in gastric cancer tissues and are associated with poor prognosis in patients with gastric cancer.2.The expression levels of HMGB1 and NF-κB were positively correlated with tumor size,depth of tumor invasion,lymph node metastasis,TNM stage and degree of tissue differentiation,but not with gender,age and distant metastasis of patients.3.HMGB1 is related to the expression of NF-κB in gastric cancer,and inhibition of HMGB1 by glycyrrhizic acid can down-regulate the expression of NF-κB,which may simultaneously promote the occurrence and development of gastric cancer.4.HMGB1 and NF-κB can promote the proliferation,migration and invasion of gastric cancer cells,and the proliferation and migration of gastric cancer cells decreased significantly after the addition of glycyrrhizic acid to inhibit the expression of HMGB1. |
