Objective Effects of L-arabinose on breast cancer cells’ invasion,metastasis,proliferation,and autophagy in vitro and in vivo,as well as the associated processes.Methods 1.L-arabinose is used to treat breast cancer cells at various doses(25mM,50 mM,100 mM),and to evaluate the activity of breast cancer cells,the CCK-8 method is employed.Using a plate cloning experiment,the effects of various L-arabinose concentrations on the growth of breast cancer cells were investigated;2.Using a Western blot,it was possible to monitor changes in the expression of EMT-related proteins such E-cadherin,N-cadherin,and vimentin.And then the autophagy-related proteins such as LC3 and Atg5;3.The effects of Larabinose on the migration and invasion of breast cancer cells were detected by scratch callus test and Transwell chamber test;4.L-arabinose,autophagy inhibitor(Baf-A1),EMT inducer(TGF-1)and Wnt/-Catenin pathway agonist(LiCl)were used to treat breast cancer cells,and Western blot was used to detect alterations in the protein expression of such Wnt/β-Catenin signaling pathway.5.Construct tumor bearing model to verify whether L-arabinose has inhibitory effect on breast cancer cells in vivo;Tissue proteins were extracted,and Western blot was utilized to determine which proteins were expressed during EMT.Results 1.Through CCK-8 detection method and plate cloning experiment,it can be concluded that L-arabinose can inhibit the proliferation of breast cancer cells;2.L-arabinose can induce autophagy in breast cancer cells,and reverse the effect of autophagy inhibitor Baf A1 on autophagy related protein expression;3.L-arabinose can inhibit the occurrence of EMT in breast cancer cells,and reverse the EMT inducer TGF-β 1.Enhancement of EMT;4.Larabinose can prevent the Wnt/-Catenin pathway from being activated in breast cancer cells and can counteract the agonist-induced activation of the pathway by LiCl;5.In nude mice tumorigenesis experiment,L-arabinose inhibited the proliferation and EMT of breast cancer cells in vivo.Conclusion L-arabinose inhibited the proliferation of breast cancer in vivo and in vitro;Larabinose inhibits EMT by inducing autophagy insertion,altering the expression of EMTrelated proteins,and preventing breast cancer cells from metastasizing;L-arabinose may pass through Wnt/ β-Catenin pathway inhibits EMT.
