Objective:The purpose of this study was to investigate the effects of Chinese medicine wogonin on the proliferation,migration and invasion of human breast cancer cells(MDA-MB-231,MCF-7)in vitro and to explore possible mechanisms of action.Methods:In this study,the representative human breast cancer cell lines MDA-MB-231 and MCF-7 were selected as the research objects.Different concentrations of wogonin were selected to act on breast cancer cells.After culture for a period of time(24h,48h,72h),MTS colorimetric method was used to map the proliferation curve of breast cancer cells,calculate the inhibition rate of the drug and the half inhibitory concentration(IC50)of the drug,and detect the effect of wogonin on the proliferation of human breast cancer cells in vitro;Cell cloning experiments were performed to examine the effects of wogonin on the colony forming ability of MDA-MB-231 and MCF-7 cells in vitro;Transwell chamber assay to examine the effects of wogonin on the migration and invasion of MDA-MB-231 and MCF-7 cells in vitro;The cell scratch test further tested the effect of wogonin on the migration ability of human breast cancer cells;Western blot qRT-PCR were used to detect the effects of wogonin on the expression of TERT(telomerase reverse transcriptase)protein and mRNA in MDA-MB-231 and MCF-7 human breast cancer cells.Western blot and qRT-PCR were used to detect the effect of wogonin on the expression of Wnt/β-catenin signaling protein and mRNA in breast cancer cells.Results:1.Inhibition of human breast cancer cells(MDA-MB-231,MCF-7)proliferation in vitro;2.Wogonin can inhibit the migration and invasion of human breast cancer cells MDA-MB-231;Human breast cancer cell MCF-7 does not have the ability to migrate/invade itself;4.The mechanism by which wogonin exerts its anti-cancer effect in breast cancer cells is related to the canonical Wnt/β-catenin signaling pathway;It can reduce the expression of TERT in human breast cancer cells and inhibit the growth of breast cancer cells.Conclusion,wogonin has a good inhibitory effect on proliferation,migration and invasion of MDA-MB-231 and MCF-7 human breast cancer cells at the in vitro cell level,and exerts its anti-cancer effect through the classical Wnt/β-catenin signaling pathway.
