Study On The Mechanism Of Jiawei Shengji Yuhong Ointment On NF-κB Signaling Pathway And VEGF Expression In Wound Tissue Of Rats With Damp-Heat Anal Fistula After Operation

Objective This study mainly discusses the effect of Jiawei Shengji Yuhong Ointment on the expression of NF-κBp65 / IL-1β / VEGF protein and its relationship with the specific molecular mechanism of promoting wound healing after anal fistula surgery.In order to solve the problem of slow wound healing after anal fistula surgery,this focus of clinicians ’attention provides a scientific theoretical basis for the development of traditional Chinese medicine.Method According to the random number table method,70 rats were divided into blank control group(blank group),vaseline group(model group),Kangfuxin solution group(control group),Jiawei Shengji Yuhong ointment group(experimental group),PDTC intervention group divided into three groups: PDTC+vaseline group(PDTC+model group),PDTC+Kangfuxin solution group(PDTC+control group),PDTC+Jiawei Shengji Yuhong ointment group(PDTC+experimental group).In addition to the blank group,the other groups were modeled with damp-heat and infectious wounds after anal fistula surgery.After successful modeling,different drug interventions were performed.At the same time,PDTC intervention group was given intraperitoneal injection of NF-κ B signaling pathway inhibitor(ammonium pyrrolidine dithiocarbamate)for 5 consecutive days.The general and wound healing of rats in each group were recorded,and the wound healing rate of rats in each group was calculated.On the 3rd,7th and 14 th day,the granulation tissue of rats in each group was taken for immunohistochemistry to detect the protein expression and tissue distribution of NF-κBp65 in wound tissue.Western blotting was used to detect the total protein content of NF-κBp65 in wound granulation tissue of rats.The expression of IL-1β/VEGF in granulation tissue was detected by ELISA.At the same time,HE staining was used for histomorphological observation.Result(1)After drug intervention,the wound healing rate of the experimental group and the PDTC+experimental group was significantly faster than that of the other groups.By 21 days,the experimental group and the PDTC+experimental group were close to complete healing,and some of them were covered with rat hair.(2)Wound healing time and healing rate : After drug intervention,compared with other groups,the experimental group and PDTC + experimental group could significantly shorten the wound healing time.The wound healing time of PDTC + experimental group was the shortest,and the difference was statistically significant(P<0.05).On the 7th and 14 th day of drug intervention,the wound healing rate of PDTC+experimental group was higher than that of the other groups(P<0.05).(3)HE staining : On the third day of drug intervention,compared with the other groups,PDTC+experimental group,experimental group and PDTC+control group had less inflammatory cell exudation,and a large number of new capillaries and scattered fibroblasts were observed;After 7 and 14 days of drug intervention,the inflammatory cells in PDTC+experimental group and experimental group gradually disappeared,and the fibroblasts gradually became collagen fibers arranged neatly and densely.(4)ELISA results : The content of IL-1β in each group decreased gradually on the 3rd,7th and 14 th day of treatment.On the 3rd day of drug intervention,there was no significant difference in the expression of IL-1β between PDTC+experimental group and PDTC+control group(P > 0.05),but it was lower than other groups(P < 0.05);On the 7th day of drug intervention,the content of IL-1 β in PDTC+experimental group was the lowest(P <0.05);On the 14 th day of drug intervention,the content of IL-1β in PDTC+experimental group and PDTC+control group was the lowest,but lower than that in the other groups(P< 0.05).On the 3rd,7th and 14 th day of treatment,the content of VEGF in each group showed a gradual increasing trend;On the 3rd day of drug intervention,the expression level of VEGF protein in the model group was the lowest(P<0.05);There was no significant difference in VEGF protein expression between PDTC+experimental group and experimental group,PDTC+control group and control group(P>0.05),but it was higher than the other groups(P<0.05),but higher than the other groups(P<0.05);On the 14 th day of drug intervention,there was no significant difference in VEGF protein expression between PDTC+experimental group and experimental group(P > 0.05),but higher than other groups(P<0.05).(5)WB results : On the 3rd day of drug intervention,the expression of NF-κBp65 protein reached the peak,and the content of model group was the highest(P<0.05).The content of NF-κBp65 in PDTC+experimental group was the lowest(P<0.05).There was no significant difference in the expression of NF-κBp65 protein between the experimental group and the PDTC+control group(P>0.05),but it was lower than the other groups(P < 0.05),but lower than the other groups(P < 0.05).(6)Immunohistochemical results : On the 3rd day of drug intervention,the protein content of NF-κBp65 in PDTC+experimental group was the lowest(P<0.05).On the 7th day of drug intervention,there was no significant difference in the expression of NF-κ Bp65 protein between PDTC+experimental group and PDTC+control group(P>0.05),but lower than the other groups(P<0.05).Conclusion This study confirmed the efficacy of Jiawei Shengji Yuhong Ointment in removing dampness and clearing heat,removing slough and promoting granulation.It can inhibit the expression of NF-κ Bp65 and IL-1β protein through NF-κ B signaling pathway,and can up-regulate the expression of VEGF protein,promote the growth of vascular endothelial cells,reduce inflammation,restore the normal function of dermal fibrous tissue,accelerate wound healing,and shorten wound healing time after anal fistula surgery.
