Part 1 Mediated Foxp3 gene silencing to enhance the cytokines and inhibit the proliferation and invasion on human periodontal ligament cell under inflammatory environmentObjective The purpose of this study was to clarify the effect of the Forkhead box P3(Foxp3)silencing on the expression of inflammatory cytokines in human periodontal ligament cell under inflammatory environment,as well as the effect on cell proliferation and invasiveness and to explore the role of Foxp3 gene in the development of periodontitis.Methods An siRNA construct specific for Foxp3 was transfected into h PDLFs.The Foxp3 silencing efficiency was verified by RT-PCR and Western Blot,and the siRNA with the best silencing effect of Foxp3 gene was screened.Using lipopolysaccharides(LPS)to simulate an under inflammatory environment in vitro,CCK-8 detected the effect of silencing Foxp3 on h PDLFs proliferation under inflammatory conditions;Wound healing experiments and Transwell assay to detect the effect of silencing Foxp3 on h PDLFs migration under inflammatory conditions;The expression inflammatory cytokines interleukin-8(IL-8)and interleukin-6(IL-6)was detected by RT-PCR and Western Blot under non-inflammatory and inflammatory conditions.Result At 24 h after siRNA transfection,RT-PCR and Western blot detection showed that the expression of Foxp3 m RNA in the si3 group decreased significantly(P<0.001),and the protein expression of Foxp3 also decreased significantly(P<0.001).Under the inflammatory environment,Foxp3 gene silencing has no significant effect on the proliferation of h PDLFs(P>0.05);Wound healing experiments and Transwell experiments showed that in an inflammatory environment,Foxp3 gene silencing promoted the migration of h PDLFs(P<0.05);Under the inflammatory environment,the expression of inflammatory factors IL-6 and IL-8 expressed by h PDLFs after the silence of the Foxp3 gene increased(P<0.05).Conclusion Under the inflammatory environment,Foxp3 gene silencing promotes the migration of h PDLFs,but has no significant effect on the proliferation of h PDLFs;The expression of inflammatory factors expressed in h PDLFs rises after the silencing of the Foxp3 gene,and the Foxp3 gene has the effect of inhibiting inflammation in periodontitis.Part 2 Case presentation: Application of Periodontal Endoscopy in Periodontal Initial TherapyObjective: To demonstrate the sequential treatment of periodontitis patients assisted by periodontoscopy,and to explore the treatmetnt effect of periodontal endoscopy.Methods: In this study,5 patients with periodontitis were subject to stage Ⅲ.-IV.of the 2018 new classification of periodontology,related contraindications were excluded,and periodontoscope-assisted periodontal sequence treatment was performed on the patients after obtaining informed consent.The periodontal condition of the patient is evaluated by periodontal examination,imaging examination,etc.After completing the basic treatment,a review was carried out 3-12 months to observe the clinical indices such as periodontal depth(PD),bleeding on probing(BOP),tooth mobility(TM)and other clinical indices after treatment to evaluate the treatment effect.Results: After receiving periodontal sequence treatment,the periodontal condition of the five patients in this study improved,the proportion of gum bleeding decreased,the depth of periodontal exploration decreased significantly,and the inflammation was improved.
