Objective:1.To determine the influences of emodin on human periodontal ligament cells (HPDLCs)in primary culture.2.To determine the effects of emodin on the expressions of IL-6 and BGP in periodontal tissues of rat periodontitis model.Methods:1.A series of experiments were performed to study the effects of emodin on the bioactivities of HPDLCs in vitro.Cell growth assay was used to evaluate the biological features of HPDLCs.Cell proliferation was determined using MTT assay,the total protein synthesis was detected by Coumassie Bright Blue method and the alkaline phosphatase activities were measured by spectrophotometric assay.2.Based on the successful rat periodontitis model,the experimental rats were randomly divided into different groups followed by oral treatment with emodin for 1,2,3,4 weeks and then the rats were sacrificed and analyzed.Pathological assay and HE staining were used to detect the general conditions and pathological changes of rat periodontal tissues. And immunohistochemical staining was conducted to determine the expressions of IL-6 and BGP in periodontal tissues of rats.Result:1.The effect of emodin on the proliferation of primarily cultured HPDLCs: Compared to the control group,the proliferative abilities of HPDLCs were increased significantly when treated with 20,50,100μg/ml of emodin for 24 hours,10,20,50μg/ml of emodin for 48 hours and 10,20,50μg/ml of emodin for 72 hours(P<0.05). 2.The effect of emodin on the synthesis of total proteins:Compared to the control group,the total amount of proteins of HPDLCs treated with 10 20,50μg/ml of emodin for 72 hours was increased dramatically(P<0.05).3.The effect of emodin on the ALP activity of HPDLCs:Compared to the control group,ALP activities of HPDLCs treated with 50μg/ml of emodin for 72 hours were enhanced significantly(P<0.05).4.The levels of IL-6 in the periodontitis tissues of periodontitis group were significantly higher than that in the normal group.Treatment with emodin decreased the levels of IL-6 in periodontitis tissues(P<0.05). The levels of BGP in the periodontitis tissues of perio- dontitis group were significantly lower than that in the normal group.Treatment with emodin increased the levels of BGP in periodontitis tissues(P<0.05). Moreover,the general conditions and pathological changes in the normal group and groups treated with emodin were much better than that in periodontitis groups.Conclusion:Emodin at the dosages ranging from 20 to 50μg/ml promoted cells proliferation and the total amount of proteins and ALP activity of HPDLCs. Also it inhibited the expression of IL-6 and increased the expression of BGP in periodontal tissues in rats periodontitis model and promoted the regeneration of the periodontal tissues.In a word,our studies suggested that emodin may have potential clinical application for periodontitis.
