Objectiveto provide clinical doctors for the general treatment of colorectal cancer clinical drug choice,for clinical practice of appropriate Chinese medicine and proprietary Chinese medicine recommended,through the network pharmacology summarizes the core of compound treatment of colorectal cancer drug mechanism,for the scholars of clinical or experimental research to provide relevant evidence,and provide ideas for the design of drug research and development.MethodsCollect the relevant clinical literature database from the establishment of CNKI,VIP,Wanfang Data and CBM to December 31,2022,screen the articles by the exclusion standard,establish the database of the extracted literature using EXCEL.Conduct data mining methods such as frequency statistics,association rules and cluster analysis through SPSS Statistics 26.0 and SPSS Mo Lder 18.0 software,and analyze the next network pharmacology analysis.In the cluster recipe that will be obtained All drugs are entered into TCMSP to obtain the target information of TCM active ingredients and active ingredients.And obtain all protein target information,these target genes were imported into Cytoscape3.9.0 software for network map of active component-target genes.After obtaining the disease targets of colorectal cancer,all the protein target information and the key disease targets were input into the Venny website for visual processing to obtain the intersection targets of drugs and disease.The PPI protein interaction network map was drawn using the STRING database and Cytoscape3.9.0.The intersection target site is then imported into the Metascape GO and KEGG data were collected from the ID database,and the top 20 results were imported into the wechat website for visual analysis of GO-BP,GO-CC,GO-MF and KEGG.ResultsThrough the screening of the literature,153 documents were included,164 prescriptions were included,and 210 drugs were counted,including Atractylodes Poria,Poria cocos,astragalus membranaceus,Codonopsis sinensis,licorice root,coix seed,white flower herb,tangerine peel,half branch lotus,and Chinese yam.The drug with the highest support is atractylodes-poria。It is concluded that the 6 groups are: cluster prescription 1 for tangerine peel,pinellia,licorice,amomum,atractylodes,poria cocos,dangshen,yam,and chicken gold;cluster prescription 2 for malt,grain bud,zedoary,and Radix Radix;cluste formula 3,respectively Astragalus;cluster prescription 4 is defeated sauce grass,rattan pear root,aurantii and coix seed;cluster prescription 5 is white peony root,Angelica,woody incense,and magnolia officinalis;cluster prescription 6 is privet,psoral,and barbary wolfberry seed.A total of 155 active components,463 standard drug targets,and 63 intersection targets were collected according to the TCMSP and HERB database.The possible roles are quercetin,licorice chalkone A,kaempferol,saponin,anthocyanin,luteolin,naringenin,soryone,mannitol,etc.In GO and KEGG analysis,the top biological processes are the negative regulation of cell apoptosis process and the positive modulation of gene expression Control,positive regulation of cell proliferation,positive regulation of protein phosphorylation,response to drugs,etc.Among the cell components are macromolecular complex,receptor complex,membrane raft,nucleus,intercellular compartment,cytoplasm,cytoplasm and nucleoplasm;molecular functions are binding enzyme,protein binding,protein kinase activity,protein tyrosine kinase activity,ubiquitin protein ligase binding,etc.Drugs may act through cancer pathways,bladder cancer,proteoglycans in cancer,EGFR tyrosine kinase inhibitor resistance,endocrine resistance,pancreatic cancer,prostate cancer,etc.ConclusionAccording to the drug cluster analysis,with the treatment principle of invigorating qi and clearing heat and detoxification,the commonly used drugs to treat colorectal cancer include atractylodes,Poria,astragalus,Dangshen,licorice,coix seed,white flower,tangerine peel,lotus,yam and so on.Cluster formula of the highest credibility,visualization for sweet sand six gentleman soup formula,through network pharmacological analysis,clustering formula through quercetin,licorice chalcone A,kaempferol,saponin,stalk,such as active ingredients,affect cancer pathway,bladder cancer,cancer proteoglycan,EGFR tyrosine kinase inhibitor resistance signaling pathway,involved in the inhibition of TP53,MYC、CCND1、STAT3、EGFR and other targets,so as to promote cell apoptosis,arrest cell cycle,degrade proteins,and reduce cell proliferation and invasion.
