BackgroundEndometrial carcinoma ranks second in the incidence rate of female malignant tumors.Its strong ability to metastasize and invasion is the main factor affecting the long-term survival rate of patients.The tumors of patients with advanced endometrial cancer have invaded or metastasized to other sites,although progress has been made in the treatment of endometrial cancer,but the prognosis of these patients is poor.Related studies have shown that the expression of long non-coding RNA(lnc RNAs)is up-regulated or down-regulated in cancer,which plays an important role in many pathways.At present,long non-coding RNA 01194(linc01194)has not been studied in endometrial carcinoma.In a word,finding and formulating effective early prognostic indicators are of great clinical significance for endometrial cancer.Methods1.The expression of linc01194 in endometrial carcinoma tissue was detected by using The Cancer Genome Atlas(TCGA)database and star Base database.Use the star Base database to find the downstream protein of linc01194.2.The expression of linc01194 in endometrial carcinoma cell lines(Ishikawa,HEC-1B,HEC-1A,KLE)and tissue samples(including 99 patients with EC and 28 patients without EC)were detected by real-time quantitative reverse-transcription polymerase chain reaction(q RT-PCR).3.siRNA gene knockout technology: culture Ishikawa cells,and when the cells are in good condition and in the logarithmic growth phase,use Lipofectamine 3000 to transfect si-linc01194 to build a linc01194 knockout cell library.4.CCK-8,wound healing,transwell test and apoptosis test were used to understand the biological function of linc01194 in endometrial carcinoma cell line.5.RNA-binding protein immunoprecipitation(RIP)was used to analyze the relationship between linc01194 and Insulin-like growth factor 2 m RNA-binding protein 1(IGF2BP1).6.The relationship between the level of linc01194 and the expression level of IGF2BP1 and sex determining region Y-box 2(SOX2)proteins was analysed through western blot.7.Data analysis: the software Graphpad Prism 8.0 was used for drawing and statistical analysis,where p < 0.05 was considered statistically significant.Results1.The expression of linc01194 is higher in endometrial carcinoma;2.Low level of linc01194 can inhibit the cell proliferation,migration,invasion and promote apoptosis;3.Linc01194 can bind to IGF2BP1 and regulate expression of IGF2BP1;4.Previous studies have shown that IGF2BP1 can promote the stability of SOX2 m RNA through m6 A modification and then promote the expression of SOX2 in endometrial carcinoma.In this study,low level of linc01194 inhibited the expression of SOX2.ConclusionsLinc01194 can promote the expression of downstream protein SOX2 through binding to IGF2BP1,thus promoting the development of endometrial carcinoma.Therefore,linc01194 may become a new biomarker for early diagnosis and prognosis prediction of endometrial carcinoma.
