Alzheimer’s disease(AD)is a multifactorial,progressive neurological disease with a large harm and a wide range,especially in the elderly.But in recent years,the survey has found that AD is showing a "younger" trend.At present,there is still no complete cure for AD,and the study of its effective therapeutic drugs is still difficult to overcome.In this context,it is necessary to develop drugs related to treating AD from a new perspective.In order to break through the bottleneck of AD drug research and development,this paper adopts a new research materials,namely with the artificial cultivation of meat ganoderma lucidum as the main body,and the laboratory provides probiotics of 2:1 new biological compound,the compound at 0.3 mg/m L when the inhibition of CL4176 C.elegans Aβ toxicity effect is good,the C.elegans median paralysis time extended by about 11.26%.Compound preparation will not inhibit the growth state of C.elegans,without toxic side effects on their reproduction and body length,but can prolong the life span of C.elegans.It can significantly reduce the fat content and ROS level in the C.elegans,that is,reduce the fat content in the late adult stage,and can reduce the production of reactive oxygen species by 33.1% in the C.elegans.For the action pathway of compound preparation,it is found that the effect of compound preparation mainly depends on the effect of microflora and has a certain relationship with metabolites.After the analysis of intestinal flora,it was found that the compound preparation could improve the abundance of beneficial bacteria in the intestine,such as Firmicutes and Actinobacteria.The results suggested that the action path of the compound preparation may be to change the composition of intestinal flora,then affect the brain-gut axis,and finally improve the AD symptoms.The results of transcriptomic analysis show that the compound preparation mainly acts through the ribosome and drug metabolism P450 related pathway,which is embodied in the influence of lysosomal content and oxidative metabolism process in cells,which is also consistent with the conclusion that the compound preparation reduces the ROS level in vivo.Further studies found that the compound preparation was able to significantly increase the expression of hsp-16.2 gene,subsequently,inhibit β amyloid aggregation,reduce neurotoxicity and improve AD symptoms.The biological compound preparation produced in this study has proved to have good anti-AD effect,which may provide a new development direction and ideas for AD drug development.
