| Meat-like Ganoderma lucidum,also known as“Tai Sui”,is a special structural component of the material with a broad development background.In this paper,the tissue structure components of Changbai Mountain Meat-like Ganoderma lucidum were obtained by high throughput screening and sequencing,and the beneficial strains of Changbai Mountain Meat-like Ganoderma lucidum were determined.The polysaccharide of Meat-like Ganoderma lucidum from Changbai Mountain was extracted by ultrasonic assisted extraction,and the antioxidant analysis was carried out based on the polysaccharide.HSCCC separation and a purification system are used for separation and purification.The extract of Meat-like Ganoderma lucidum from Changbai Mountain was cultured in solid,semi-solid and liquid media through biological medium,and the tissue differentiation was investigated.The research is as follows:The single factor experiment was used to determine the influencing factors for the extraction of polysaccharides from Meat-like Ganoderma lucidum in Changbai Mountain.The ultrasonic extraction process was optimized by response surface methodology,and the extracted polysaccharides were separated and purified.The antioxidant analysis was performed after separation and purification.The results were as follows:extraction temperature 51.63℃,ultrasonic time 4.11h,ultrasonic power 286.32W,solid-liquid ratio1:31.89g/ml.Under these conditions,the maximum extraction rate of Meat-like Ganoderma lucidum water extract from Changbai Mountain was 10.2796%.The HSCCC separation and purification system was used for separation and purification.Within 230 min of separation and purification,the peaks appeared at 110 min,125 min and 205 min,respectively,and were named as CBG1,CBG2and CBG3.The preliminary separation and purification products were obtained.The determination of reducing power and free radical scavenging experiments showed that Meat-like Ganoderma lucidum polysaccharides from Changbai Mountain had obvious antioxidant capacity,and CBG3had the strongest ability and CBG1had the weakest ability.The high-throughput sequencing technology was used to determine the microorganisms in Meat-like Ganoderma lucidum of Changbai Mountain and construct the anti-inflammatory model.The cell survival rate was detected by MTT method for anti-inflammatory analysis.The results were as follows:Meat-like Ganoderma lucidum microorganisms in Changbai Mountain were divided into bacterial diversity fungal diversity,and the main culturable substances of bacterial diversity were Lachnospiraceae,Romboutsia,Royal Lactobacillus,and Caulobacter.The main culturable substances of fungal diversity are Fusarium and Hannaella.At different concentrations,the cell survival rate was maintained at more than 90%,indicating that the screening microorganisms of Meat-like Ganoderma lucidum from Changbai Mountain had no toxic effect on cells.The release of NO was measured by Griess,and the release of IL-6,IL-1βand PGE2was detected by ELISA.The extract had a significant inhibitory effect on the release level of the existing model(P<0.05).The experiment showed that the extract of Meat-like Ganoderma lucidum from Changbai Mountain could reduce inflammation and play an anti-inflammatory role.Through the study of tissue culture in biological medium,solid,semi-solid and liquid media were used.The tissue culture analysis of Meat-like Ganoderma lucidum showed that the mycelium division of Meat-like Ganoderma lucidum was not obvious,the growth was slow,and the cultivation cycle of Changbai Mountain Meat-like Ganoderma lucidum was long. |
