Differential Expression Of Actin Fiber-associated Protein 1-antisense RNA1 In Ovarian Endometriosis Stage Ⅲ And Ⅳ And Its Clinical Significance

Objective:In this study,long non-coding RNA actin filament-associated protein 1 antisense RNA1(LncRNA AFAP1-AS1)was measured in patients with ovarian endometriosis(OEC)at stage Ⅲ and Ⅳ(according to rASMR score).The expression levels of LncRNA AFAP1-AS1 in lesion tissue of OEC patients and the clinical indicators of OEC patients were discussed in patients requiring surgical treatment,including the expression levels of ectopic lesion tissue,adjacent lesion tissue,in-place endometrial tissue and normal endometrial tissue.Namely,whether there is a correlation between clinical pain score(VAS)and fertility index score(EFI).Methods:LncRNA was determined by real-time quantitative fluorescence qRT-PCR and immunohistochemistry The expression levels of AFAP1-AS1 in ectopic lesion tissue,para-lesion tissue,in-place endometrial tissue and 47 normal endometrial tissue of 47 OEC patients requiring surgical treatment at Stage Ⅲ and Ⅳ(rASMR stage)were collected.Meanwhile,general clinical data of the above 47 patients with OEC were collected.Preoperative VAS,intraoperative EFI,postoperative gonadotropin-releasing hormone agonist drug therapy(GnRH-a injection)and natural pregnancy within 2 years after surgery were used to divide 47 patients with OEC into two groups according to their pregnancy status:Pregnant group(n=31)and non-pregnant group(n=16),47 patients were divided into two groups according to rASMR score:Stage Ⅲ patients(n=29);Stage Ⅳ patients(n=18).SPSS26.0 statistical software was used to compare the differences of age,infertility time,infertility type,postoperative GnRH-a injection,preoperative VAS,intraoperative EFI and clinical stage of OEC patients(according to rASMR stage)between the pregnant group and non-pregnant group.And the difference of preoperative VAS and intraoperative EFI between stage Ⅲ andⅣ patients with OEC was compared.The expression level of LncRNA AF AP1-AS 1 in focal tissues of patients with stage Ⅲ and Ⅳ OEC was taken as the abscissa,and the preoperative VAS and intraoperative EFI of patients with stage Ⅲ and Ⅳ were taken as the ordinate to draw the scatterplot to analyze whether there was any correlation between them.Results:1.In this study,qRT-PCR and immunohistochemistry were used to determine the expression levels of LncRNA AFAP1-AS1 in stage Ⅳ lesion tissue,adjacent lesion tissue,and in-place endometrial tissue of patients with OEC were higher than those in stage Ⅲ lesion tissue,adjacent lesion tissue,and in-place endometrial tissue(P<0.05),and LncRNA AFAP1-AS1 expression was higher in ectopic lesion tissue than in normal endometrial tissue(P<0.05).2.A total of 47 patients with OEC were included in this study.Within 2 years after laparoscopic surgery,31 patients had a successful natural pregnancy and 16 patients did not have a pregnancy,with a pregnancy rate of 65.96%.Among the 33 patients with infertility associated with OEC,19 patients had a successful natural pregnancy and 14 patients still did not have a pregnancy,with a pregnancy rate of 57.57%.3.Postoperative pregnancy in patients with OEC may be correlated with age,rASMR score,infertility duration,infertility type,postoperative G-nRHa injection,and intraoperative EFI(P<0.05)and had no correlation with preoperative VAS(P>0.05).4.Clinical stage Ⅲ and Ⅳ of patients with OEC(according to rASMR stage)were not correlated with preoperative VAS(P>0.05),correlated with EFI(P<0.05).5.There was no correlation between the expression level of LncRNA AFAP1-AS1 in focal tissues of patients with OEC and preoperative VAS(correlation coefficient r=0.10506,P=0.59>0.05;Ⅳ phase correlation coefficient r=0.17355,P=0.49>0.05),and was negatively correlated with EFI(intermediate Ⅲ correlation coefficient r=-0.78376,P=0.000<0.001;Ⅳ phase correlation coefficient r=-0.74173,P=0.000<0.001).Conclusions:LncRNA AFAP1-AS 1 was differentially expressed in phase Ⅲ and Ⅳfocal tissues,para-focal tissues,and in-place endometrial tissues of patients with OEC.The expression level of LncRNA AFAP1-AS1 was not correlated with preoperative VAS of patients with OEC,but was negatively correlated with intraoperative EFI.
