Background:Renal cell carcinoma is one of the most common malignant tumors of urinary system in the world.It is the most common solid lesion of kidney.Its morbidity and mortality are high,and it has posed a serious threat to human health.At present,surgical therapy,targeted therapy and immunotherapy are the main therapeutic methods for renal cell carcinoma.However,since most patients have been diagnosed in the middle and advanced stage,surgical therapy cannot achieve good efficacy.However,although the efficacy of targeted therapy and immunotherapy is good,there are drug resistance,side effects and expensive problems.Therefore,it is urgent to find safe and effective methods to prevent and treat renal cell carcinoma.Aspirin is a common non-steroidal anti-inflammatory drug,which is mainly used in clinical treatment of cold,fever,headache and prevention of cardiovascular and cerebrovascular diseases.In recent years,a large number of studies have found that aspirin also plays an important role in anti-tumor,and its safety is higher than some anti-tumor drugs.Studies have reported that regular aspirin use can improve the survival rate of patients with colorectal,liver,and stomach cancers.However,there is no relevant report on the effect of aspirin on renal cell carcinoma and its mechanism of action.This paper will analyze the effect of aspirin on renal cell carcinoma and further explore the mechanism of aspirin’s regulation on renal cell carcinoma.Objective:1、To confirm whether aspirin can inhibit the proliferation and migration of renal cancer cells and promote apoptosis;2、To verify the regulatory effects of aspirin on proliferation,migration and apoptosis of renal cancer cells through autophagy.Methods:1、 The proliferative ability of renal carcinoma cells was detected by CCK-8 assay and plate cloning assay;2、The migration level of renal carcinoma cells was detected by Transwell test and scratch test;3、Apoptosis of renal carcinoma cells was detected by Annexin V/PI assay and Western blot;4、The expressions of autophagy-related proteins in renal carcinoma cells were detected by Western blot.Results:In this study,two types of renal carcinoma cell lines(ACHN and 786-O cells)were selected as the research objects.Firstly,the effect of aspirin on the proliferation of renal carcinoma cells was detected by CCK-8 and plate cloning experiments.The results showed that aspirin intervention could significantly inhibit the survival rate of ACHN and 786-O cells compared with the normal control group.The cell survival rate was negatively correlated with aspirin concentration and treatment time(P<0.05).Transwell and scratch experiments showed that aspirin significantly inhibited the proliferation and migration of renal carcinoma cells compared with the control group(P<0.01).In addition,Annexin V/PI assay showed the apoptosis of kidney cancer cells was significantly increased after aspirin treatment,and the expression of apoptosis-related protein Bcl-2 was significantly decreased and the expression of Bax protein was upregulated.Meanwhile,compared with blank control group,aspirin treatment significantly promoted the expressions of autophagy related proteins LC3 B and Beclin-1,and inhibited the expression of p62(P<0.05).Compared with chloroquine treatment group,aspirin + chloroquine group significantly promoted the expression of LC3 B and inhibited the expression of p62(P<0.05).Conclusion:1、Aspirin can significantly inhibit the proliferation and migration of human renal carcinoma cell lines ACHN and 786-O,and promote their apoptosis;2、Aspirin can affect the function of kidney cancer cells by enhancing autophagy. |
