Objective:Since the 21 st century,with the continuous deepening of research on metformin drugs,the medicinal value of metformin is no longer limited to hypoglycemic efficacy,and many of its drug reuse potential has been continuously explored.Due to the particularity of the pharmaceutical industry,medical data resources,while having high value,also exhibit characteristics such as incompleteness,non standardization,and redundancy.Therefore,exploring the thematic evolution of metformin drug research,revealing the research overview,therapeutic use changes,research hotspot changes,and exploring the reuse potential of metformin drugs in the future have certain guiding significance for medical researchers to quickly grasp the development trends of disciplines and make medical strategic decisions.Methods : This study selected all the literature related to metformin drugs included in the Medline database Ebsco version from 1959 to 2020 as the data source.The research object is subject words,sub subject words,tag words,and index words.Using tools such as Excel,Tableau,SPSS,and g CLUTO,the evolution analysis of metformin drug research based on the characteristics of network structure and the evolution analysis of metformin drug research based on the characteristics of literature content are conducted from the perspectives of structure and content.The research overview,research hotspots,and changes in therapeutic uses of metformin drugs in different periods have been revealed from macro and micro perspectives.Result:Through statistical analysis,it was found that the research on metformin drugs started around 1960.Around 1970,research on metformin drugs fell into a low ebb.It was not until 1997 that metformin drugs were re used that they entered a golden period of development;Based on the evolution analysis of metformin drug research based on the characteristics of network structure,it is found that the research scale of metformin drug research is continuously expanding,the degree of agglomeration is continuously decreasing,the research topics are differentiated,and the depth and breadth of research content and research are also increasing;Based on the analysis of the evolution of research on metformin drugs based on the characteristics of literature content,it was found that the research topics of metformin drugs in the treatment of obesity,cardiovascular and cerebrovascular diseases,polycystic ovary syndrome,anticancer,anti-inflammatory,regulating intestinal microbiome,and delaying aging,in addition to their hypoglycemic effects,have new value in the treatment of obesity,cardiovascular and cerebrovascular diseases Neurodegenerative diseases and androgen alopecia have potential for drug reuse.Conclusion:Through the analysis of thematic evolution in the field of metformin drugs from the perspective of structure content,this study found that: in terms of the evolution of network structure,the overall situation of the network varies with different stages of time;In terms of the evolution of content characteristics,the changes in the research topic of therapeutic uses of metformin drugs mainly focus on the 21 st century,and there have been six significant changes in therapeutic uses from1959 to 2020.The results of cluster analysis and correspondence analysis also confirm the changing characteristics of network structure.The organic combination of social network analysis theory,topic evolution theory,and correspondence analysis theory can well reveal the research overview of metformin drugs in different periods,changes in drug therapeutic uses,research hotspots,and predict the reuse potential of metformin drugs in new research in the future.
